摘  要:目的:研究血脂异常冠心病大鼠血管内皮损伤机制。方法:根据培养方法,20只大鼠被均分为健康对照组和血脂异常冠心病组。观察比较两组培养后血脂水平、血清TNF-α、IL-1β、IL-6、SOD、GSH、MDA水平、VEGF、Bcl-2及半胱氨酸蛋白酶(Caspase-9)相对表达量。结果:与健康对照组比较,血脂异常冠心病组血清TC[(1.00±0.11)nmol/L比(6.56±0.27)nmol/L]、TG[(0.55±0.06)nmol/L比(3.31±0.18)nmol/L]、LDL-C[(0.72±0.03)nmol/L比(3.73±0.13)nmol/L]、TNF-α[(6.97±0.16)ng/L比(21.21±1.17)ng/L]、IL-1β[(3.05±0.10)ng/L比(9.96±0.46)ng/L]、IL-6[(1.54±0.11)ng/L比(16.16±0.44)ng/L]、MDA[(5.22±0.27)nmol/ml比(9.17±0.20)nmol/ml]水平、Caspase-9相对表达量[(11.86±1.21)%比(35.66±1.02)%]均显著升高,血清HDL-C[(1.10±0.11)nmol/L比(0.40±0.03)nmol/L]、SOD[(250.12±6.47)U/ml比(162.23±3.89)U/ml]、GSH水平[(217.76±4.47)IU比(165.53±4.74)IU]、VEGF[(76.07±3.43)%比(37.82±1.21)%]、Bcl-2相对表达量[(63.45±2.60)%比(10.11±0.79)%]均显著降低,P均=0.001。结论:加重的血管内皮炎症反应及升高的血清氧自由基水平会导致血脂异常冠心病的血管内皮组织损伤,下降的Bcl-2相对表达量和升高的Caspase-9相对表达量会促进血管内皮细胞凋亡。Objective:To study the mechanism of vascular endothelial injury in rats with coronary heart disease(CHD)complicated dyslipidemia.Methods:According to culture method,a total of 20 rats were divided into healthy control group and dyslipidemia complicated CHD(D-CHD)group.Levels of blood lipids,serum TNF-α,IL-1β,IL-6,SOD,GSH and MDA,relative expression amounts of VEGF,Bcl-2 and Caspase-9 were observed and compared between two groups.Results:Compared with healthy control group,there were significant rise in serum levels of TC[(1.00±0.11)nmol/L vs.(6.56±0.27)nmol/L],TG[(0.55±0.06)nmol/L vs.(3.31±0.18)nmol/L],LDL-C[(0.72±0.03)nmol/L vs.(3.73±0.13)nmol/L],TNF-α[(6.97±0.16)ng/L vs.(21.21±1.17)ng/L],IL-1β[(3.05±0.10)ng/L vs.(9.96±0.46)ng/L],IL-6[(1.54±0.11)ng/L vs.(16.16±0.44)ng/L]and MDA[(5.22±0.27)nmol/ml vs.(9.17±0.20)nmol/ml]and relative expression amount of Caspase-9[(11.86±1.21)%vs.(35.66±1.02)%]in D-CHD group,and significant reductions in serum levels of HDL-C[(1.10±0.11)nmol/L vs.(0.40±0.03)nmol/L],SOD[(250.12±6.47)U/ml vs.(162.23±3.89)U/ml]and GSH[(217.76±4.47)IU vs.(165.53±4.74)IU],relative expression amounts of VEGF[(76.07±3.43)%vs.(37.82±1.21)%]and Bcl-2[(63.45±2.60)%vs.(10.11±0.79)%]in D-CHD group,P=0.001 all.Conclusion:Aggravated vascular endothelial inflammation and increased serum oxygen radical levels can cause vascular endothelial tissue injury in CHD with dyslipidemia.Decreased relative expression amount of Bcl-2 and increased relative expression amount of Caspase-9 can promote vascular endothelial cell apoptosis.
