摘 要:目的探讨异维A酸对痤疮丙酸杆菌的体外抗菌活性及机制。方法通过生长抑制实验观察异维A酸对痤疮丙酸杆菌的抑制作用并测量最小抑菌浓度(MIC);通过对细菌细胞壁、细胞膜的研究揭示药物抑制细菌的作用机制;通过透射电镜观察细菌的形态结构变化。结果异维A酸浓度越高对痤疮丙酸杆菌抑制作用越强,最低抑菌浓度为80μg/mL。药物浓度越高,钾离子、镁离子和碱性磷酸酶(AKP)外流越明显,且随时间延长,逐渐增加。药物浓度越高,受损和死亡细菌越多,存活细菌越少,细菌细胞膜电位降低越明显,且有时间依赖性。电镜观察发现,药物浓度越高,细菌的结构破坏越明显。结论异维A酸对痤疮丙酸杆菌有直接杀灭作用,其作用机制可能与增加细胞通透性、降低细胞膜电位等,从而破坏细菌细胞结构,导致其死亡有关。Objective To explore the antibacterial activity and mechanism of isotretinoin against Propionibacterium acnes in vitro.Methods The inhibitory effect of isotretinoin on Propionibacterium acnes was observed by growth inhibition experiment,and the minimum inhibitory concentration(MIC)was measured.The mechanism of drug inhibition on bacteria was revealed through the study of cell wall and cell membrane.The morphological and structural changes of bacteria were observed by transmission electron microscope.Results The higher the concentration of acitretinoin was,the stronger the inhibition was.The higher the drug concentration,the more obvious the outflow of potassium ion,magnesium ion and alkaline phosphatase(AKP),and gradually increased with time.The higher the drug concentration,the more damaged and dead bacteria,the less viable bacteria.The higher the drug concentration,the more obvious the decrease of cell membrane potential.Electron microscopy showed that the higher the concentration of the drug,the more obvious the destruction of bacterial structure.Conclusion Isotretinoin has a direct killing effect on Propionibacterium acnes.The mechanism of action may be related to the increase of cell permeability and the decrease of cell membrane potential,thus destroying the bacterial cell structure and leading to its death.
