摘 要:目的了解凉山州健康无偿献血人群血液微生物组信息,检测血液中可能存在的病原体,并调查其流行情况。方法随机采集凉山州健康献血人群血液标本1299(人)份,提取总核酸后进行高通量测序,通过宏基因组学分析,确定微生物组。采用实时荧光定量PCR和酶联免疫吸附试验分别检测单份血清标本的HSV DNA和HSV抗体,对比分析不同性别、年龄、职业及受教育程度等献血者HSV阳性率。结果通过高通量测序,DNA pool获得数据3.49 GB,去除人类DNA背景后,结果注释为细菌、寄生虫/真菌和病毒的DNA序列片段分别是213057reads,10623reads,15reads;cDNA pool获得数据2.79 GB,去除人类DNA背景后,结果注释为细菌、寄生虫/真菌和病毒的片段分别是4105600reads,18446reads和0reads。HSV-1和HSV-2血清总抗体阳性率为8.62%(112/1299)和18.32%(238/1299),HSV DNA阳性率0.77‰(1/1299)。结论本研究结果证实了凉山州健康献血人群血液中的微生物组信息,并鉴定了其中的新发再发病原体。凉山州HSV感染具有区域特异性。鉴于HSV-2和HIV感染的协同相关性,考虑率先在凉山州以及其他HIV高流行区的不同人群中开展HSV-2流行病学调查,以期服务于HIV感染防控工作。Objective To map a comprehensive description of microbiome presented in healthy blood donations in Liangshan Yi Autonomous Prefecture,and detect the potential pathogens and its prevalence.Methods A total of 1299 blood samples were randomly selected from healthy blood donors in Liangshan Prefecture.Total nucleic acids were extracted and sequenced by high-throughput sequencing,and the microbiome was determined by metagenomics analysis.Quantitative real-time PCR(qPCR)and ELISA assays were used to detect antibodies against HSV DNA and HSA in each sample.The prevalence of HSV in healthy blood donors was compared in terms of gender,age,occupation,education level,and frequency of donation.Results 3.49 GB data were obtained from DNA pool through high-throughput sequencing.After filtering the data of human genome,the DNA sequences annotated as bacteria,parasites/fungi and viruses were 213057,10623 and 15 reads,respectively.A total of 2.79 GB data were obtained from cDNA pool,after filtering the data of human genome,the fragments annotated as bacteria,parasites/fungi and viruses were 4105600,18446 and 0 reads,respectively.The prevalence of IgG and/or IgM antibodies against herpes simplex virus type 1 and 2 were 8.62%(112/1299)and 18.32%(238/1299),and that of nucleic acid was 0.77‰(1/1299).Conclusion The microbiome of healthy blood donors in Liangshan Prefecture and the potential pathogens were identified in this study.Regional specificity of HSV infections emerged in Liangshan Prefecture.Considering the collaboration between HSV-2 and HIV infection,epidemiological investigation of HSV-2 infection should be conducted preferentially among different populations in Liangshan Prefecture and other HIV high prevalence areas in order to benefit the prevention and control of HIV.
