摘 要:本研究选取自凝集能力不同的两株汉赛巴尔通体临床分离菌株进行巨噬细胞感染实验。采用冻融法比较两种巴尔通体菌胞内存活效率,并以免疫荧光技术观察巨噬细胞内巴尔通体吞噬体的熟化差异。结果显示:弱凝集巴尔通体菌在巨噬细胞内的胞内适应性优于强凝集反应菌株,在24 h检测周期内弱凝集反应菌株的存活效率较高;强凝集反应菌株的Rab7阳性吞噬体显著高于弱凝集巴尔通体菌株;溶酶体探针检测结果显示,强凝集反应菌株吞噬体的熟化程度也高于弱凝集反应菌株。本研究结果提示自凝集反应对巴尔通体菌抵抗巨噬细胞杀伤存在负调控作用。In this study,two clinical strains of Bartonella henselae with different auto-agglutination abilities were selected for macrophage infection experiment.The intracellular survival efficiency of B.henselae was studied by freezing-thawing method and maturation of phagosomes was examined using immunofluorescence assay.The results showed that the weak agglutinating B.henselae strain ATCC 49882 showed better intracellular adaptability in macrophages than the strong agglutinating strain JK33.More ATCC 49882 bacteria survived in macrophages than JK33 in 24 hours.Moreover,more Rab7 positive phagosomes were observed in JK33 infected macrophages than ATCC 49882 infected samples.As expected,more phagosomes of JK33 were acidified than ATCC 49882.The results of this study suggested that auto-agglutination had negative effect on intracellular adaptability of B.henselae.
