摘 要:为建立一种快速灵敏检测犬细小病毒的免疫层析方法,本研究将羧基化CdSe/ZnS量子点与犬细小病毒单克隆抗体5G7共价偶联作为捕获探针,以犬细小病毒单克隆抗体8H7、羊抗鼠IgG作为检测线、质控线制备量子点免疫层析试纸条。结果显示,该试纸条灵敏度高,最低检测限为103 TCID50/mL;特异性强,与犬瘟热病毒、犬冠状病毒、犬腺病毒1型无交叉反应;稳定性好,室温密封保存30 d,其灵敏度和特异性无变化;临床样本检测,与PCR方法符合率为94%,准确率高于商品化胶体金试纸条。综上所述,该试纸条灵敏度高、特异性强、准确率高,适用于犬细小病毒病临床快速诊断。To develop a immunochromatographic strip method for rapid and sensitive detection of Canine parvovirus,carboxylated CdSe/ZnS quantum dots(QDs)were conjugated to specific monoclonal antibody 5G7 against Canine parvovirus as a capture probe.The monoclonal antibody 8H7 against Canine parvovirus and goat anti-mouse IgG were sprayed on the test line and control line.As results,the QDs strips detected Canine parvovirus with a detection limit of 103 TCID50/mL and had no cross reaction with Canine distemper virus,Canine coronavirus and Canine adenovirus type 1.The QDs strips could be stored in room temperature for 30 days.As compared with commercial colloidal gold strips,the QDs strips had higher accuracy when testing clinical samples.The coincidence rate of QDs strips was 94%with PCR.In conclusion,the developed QDs immunochromatographic strip was sensitive,specific and accurate for the detection of Canine parvovirus,which might be applicable to clinical diagnosis.
