摘 要:为有效测定大白菜中的木质素含量,本试验利用木质素乙酰化后在280 nm处具有吸收峰值的特性,建立并优化了比色法测定大白菜木质素含量的反应条件。结果表明,比色法测定大白菜木质素含量的最佳反应条件为:乙酰溴浓度30%,反应温度70℃,反应时间40 min。采用该优化方法测定了14份大白菜材料的木质素含量,并与木质素测定试剂盒检测结果进行比较,表明优化后的比色法较为准确、简便。In order to effectively determine the lignin content in Chinese cabbage,this experiment used the characteristic of lignin to have an absorption peak at 280 nm after acetylation,and established and optimized a colorimetric method to determine the reaction condition of lignin content in Chinese cabbage.The results showed that the optimal reaction conditions for the colorimetric determination of lignin content in Chinese cabbage were:30%acetyl bromide concentration,70℃reaction temperature,40 min reaction time.The lignin contents of 14 accessions of Chinese cabbage were determined by this optimized method.These results were compared with that of the test by lignin testing kit,indicating after optimization this colorimetric method was more accurate,simple and convenient.
