摘  要:目的探讨分别用来曲唑(LTZ)和脱氢表雄酮(DHEA)建立的多囊卵巢综合征(PCOS)大鼠模型的生殖和代谢异常的特征。方法6周龄SPF级SD雌性大鼠24只随机分为空白组,来曲唑组,脱氢表雄酮组,从建模前后大鼠体重增长、动情周期、卵巢及子宫组织形态学变化、胰岛素抵抗等指标对2个模型进行评价。结果与空白组相比,来曲唑组和脱氢表雄酮组体重均显著上升(均P<0.01),来曲唑组体重高于脱氢表雄酮组(P<0.01);来曲唑组和脱氢表雄酮组均失去完整动情周期;来曲唑组和脱氢表雄酮组卵巢均发生多囊样改变,脱氢表雄酮组子宫局部形态变化更明显。空白组、来曲唑组和脱氢表雄酮组卵巢湿重分别为(0.12±0.00),(0.17±0.02),(0.23±0.01)g;子宫湿重分别为(0.28±0.02),(0.17±0.02),(0.79±0.14)g;空腹血糖(FBG)分别为(4.55±0.39),(5.84±0.99),(5.35±0.93)mmol·L^(-1);空腹胰岛素(FINS)分别为(8.70±0.51),(9.60±1.28),(15.28±0.50)uU·mL^(-1);胰岛素抵抗指数(HOMA-IR)分别为1.76±0.14,2.51±0.60,3.86±0.52。空白组着床率为100%,平均着床胚泡数为12.80±0.45,而来曲唑组和脱氢表雄酮组着床率和平均着床胚泡数均为0,差异均有统计学意义(均P<0.05)。结论脱氢表雄酮造模法更符合PCOS患者的病理生理特征,是研究PCOS理想的动物模型。Objective Animal models of polycystic ovarian syndrome(PCOS)in rats were established by letrozole(LTZ)and dehydroepiandrosterone(DHEA)respectively,to discuss and compare the characteristics of abnormal reproduction and metabolism.Methods SD female rats aged six weeks were randomly divided into 3 groups:blank group,letrozole group,and dehydroepiandrosterone group.The model was evaluated from the weight gain,estrous cycle,morphological changes of ovary and uterus,and insulin resistance in rats.Results Compared with blank group,the body weight of letrozole group and dehydroepiandrosterone group were increased significantly(all P<0.01),the weight of letrozole group was higher than that of dehydroepiandrosterone group(P<0.01);both the letrozole group and the dehydroepiandrosterone group lost the complete estrus cycle;both letrozole group and dehydroepiandrosterone group had polycystic changes in the ovaries,and the local uterine morphology changes were more obvious in the dehydroepiandrosterone group.The ovarian wet weight in blank,letrozole and dehydroepiandrosterone groups were(0.12±0.00),(0.17±0.02),and(0.23±0.01)g;the uterine wet weight were(0.28±0.02),(0.17±0.02),and(0.79±0.14)g;the fast blood glucose(FBG)levels were(4.55±0.39),(5.84±0.99),and(5.35±0.93)mmol·L,the fasting insulin(FINS)levels were(8.70±0.51),(9.60±1.28),and(15.28±0.50)uU·mL^(-1);the Homeostasis model assessment of insulin resistence(HOMA-IR)were 1.76±0.14,2.51±0.60,3.86±0.52;the implantation rate in group B was 100%,and the average number of implanted blastocysts was 12.80±0.45,while the implantation rates in the letrozole group and the dehydroepiandrosterone group were similar.The average number of implanted blastocysts is 0.The differences were all statistically significant(all P<0.05).Conclusion The method of establishing a rat model of PCOS with dehydroepiandrosterone is more in line with the pathophysiological characteristics of PCOS patients,and is an ideal animal model for studying PCOS.
