摘 要:目的探究miR-136-5p在宫颈癌中的表达、功能及机制。方法常规培养人宫颈癌细胞株(C33A)。将实验分为miR-control组和miR-136-5p组。用逆转录-实时荧光定量聚合酶链反应(qRT-PCR)检测宫颈癌患者组织和细胞系中miR-136-5p以及MCM5 mRNA的表达水平。用蛋白质印迹(Western blot)法检测MCM5蛋白在宫颈癌细胞系中的表达。用CCK-8、BrdU和Transwell实验检测miR-136-5p对宫颈癌细胞增殖、迁移和侵袭的影响。通过Targetscan在线分析数据库和双荧光素酶报告基因分别预测和验证miR-136-5p和MCM5之间的靶向调控关系。结果 miR-136-5p在癌旁组织和癌组织中的表达水平分别为1.02±0.21和0.53±0.18,差异有统计学意义(P<0.05)。miR-control组和miR-136-5p组细胞增殖分别为0.65±0.06和0.43±0.04,迁移的细胞分别为65±6和40±4,侵袭的细胞数目分别为50±5和30±3,差异均有统计学意义(均P<0.05)。机制上,miR-136-5p通过靶向抑制MCM5表达,从而在宫颈癌中充当抑癌因子。结论 miR-136-5p通过负向调控MCM5的表达参与抑制宫颈癌细胞的增殖、迁移和侵袭。Objective To investigate the expression, function and mechanism of miR-136-5 p in cervical cancer(CC). Methods CC cells(C33 A) were conventionally cultured and divided into miR-control and miR-136-5 p groups.The expression levels of miR-136-5 p and MCM5 mRNA in tissues and cell lines of patients with CC were detected by qRT-PCR. Western blot was used to detect the expression of MCM5 protein in CC cell lines. CCK-8, BrdU and Transwell experiments were used to detect the effects of miR-136-5 p on the proliferation, migration and invasion of CC cells. TargetScan database and dual-luciferase reporter genes were used to predict and verify the targeted regulatory relationship between miR-136-5 p and MCM5, respectively. Results The expression of miR-136-5p in adjacent tissues and ovarian cancer tissues were 1.02±0.21 and 0.53±0.18, the difference was significant(P<0.05). The proliferation rates in miR-control group and miR-136-5 p group were 0.65±0.06 and 0.43±0.04, the number of migrated cells were 65±6 and 40±4, the number of invaded cells were 50±5 and 30±3, the difference were all significant( all P < 0. 05). Mechanistically,miR-136-5 p inhibited its expression by targeting MCM5,thereby acting as a tumor suppressor in CC. Conclusion miR-136-5 p is involved in inhibiting the proliferation,migration and invasion of CC cells by negatively regulating the expression of MCM5.
