摘　　要：目的探究西红花提取物对柯萨奇B3病毒(CVB3)感染心肌炎小鼠模型心肌细胞凋亡和心肌纤维化的影响.方法采用CVB3感染小鼠构建病毒性心肌炎模型,西红花提取物组以3 mg/kg西红花提取物灌胃处理,1次/d,连续处理7d,对照组和模型组给予等量生理盐水灌胃.药物处理7d后,采用超声评价小鼠心功能情况,观察血清心肌损伤标记物[心肌肌钙蛋白T(cTnT)、肌酸激酶同工酶(CK-MB)]和炎性因子[白细胞介素(IL)-6、-1β、肿瘤坏死因子-α(TNF-α)]水平,心肌组织苏木素-伊红(HE)染色评价病理变化、原位末端转移酶标记(TUNEL)染色检测心肌凋亡情况及马松(MASSON)染色评价纤维化水平,蛋白印迹(WB)法检测心肌组织中半胱天冬酶(caspase-3、-9)、凋亡基因Fas、转化生长因子(TGF-β1)、Smad2/3表达情况.结果与对照组比较,病毒性心肌炎小鼠血清cTnT、CK-MB水平升高(P<0.05),心率(HR)和左心室射血分数(LVEF)降低(P<0.05);与病毒性心肌炎组比较,西红花提取物组cTnT、CK-MB水平降低(P<0.05),HR、LVEF升高(P<0.05);与对照组比较,病毒性心肌炎组血清IL-6、-1β、TNF-α表达增加(P<0.05),与病毒性心肌炎组比较,西红花提取物组IL-6、-1β、TNF-α表达减少(P<0.05);与对照组比较,病毒性心肌炎组凋亡细胞数、caspase-3、caspase-9、Fas表达增加(P<0.05);与病毒性心肌炎组比较,西红花提取物组凋亡细胞数、caspase-3、caspase-9、Fas表达减少(P<0.05);与对照组比较,病毒性心肌炎组心肌组织纤维化、TGF-β1、Smad2/3磷酸化水平升高(P<0.05),与病毒性心肌炎组比较,西红花提取物组心肌组织纤维化、TGF-β1、Smad2/3磷酸化水平降低(P<0.05).结论西红花提取物通过减轻炎症反应、抑制心肌细胞凋亡和纤维化,保护CVB3感染引起的病毒性心肌炎,可能与调控心肌组织中caspase-3、caspase-9、Fas及TGF-β/Smad通路表达有关.OBJECTIVE To explore the effect of saffron extract on myocardial apoptosis and myocardial fibrosis indexes of the mice models with myocarditis caused by Coxsackie B3 virus(CVB3)infection.METHODS The mice with CVB3 infection were chosen to build the viral myocarditis models.The saffron extract group was administered intragastrically with 3 mg/kg saffron extract,once a day,for 7 consecutive days,while the control group and the model group were treated with the same volume of normal saline.After the drug treatment for 7 days,the cardiac function of the mice was evaluated by ultrasound,the levels of serum myocardial injury markers[cardiac troponin T(cTnT),creatine kinase isoenzyme(CK-MB)]and inflammatory factors[interleukin(IL-6,-1β),tumor necrosis factor-α(TNF-α)]were observed,the pathological changes were evaluated by myocardial tissue hematoxylin-eosin(HE)staining,the status of myocardial apoptosis was detected by TdT-mediated dUTP-biotin nick end labeling(TUNEL)staining,the fibrosis level was evaluated by means of MASSON staining,and the expression levels of caspases(caspase-3,-9),apoptosis gene Fas,transforming growth factor(TGF-β1)and Smad2/3 in myocardial tissues were detected by using Western blot.RESULTS The levels of serum cTnT and CK-MB of the viral myocarditis group were significantly higher than those of the control group(P<0.05),the heart rate(HR)and left ventricular ejection fraction(LVEF)of the viral myocarditis group were significantly lower than those of the control group(P<0.05).The levels of cTnT and CK-MB of the saffron extract group were significantly lower than those of the viral myocarditis group(P<0.05),while the HR and LVEF of the saffron extract group were significantly higher than those of the viral myocarditis group(P<0.05).The expression levels of serum IL-6,-1βand TNF-αof the viral myocarditis group were significantly higher than those of the control group(P<0.05);the expression levels of IL-6,-1βand TNF-αof the saffron extract group were significantly lower than those of the vi
