摘 要:目的探讨灵芝多糖(GLPs)对川崎病(KD)模型小鼠淋巴细胞转化和外周血细胞因子的调节作用.方法雄性BALB/c小鼠随机分为对照组、KD模型组、GLPs低、高剂量组(50 mg/kg、100 mg/kg)和阿司匹林组;对照组注射磷酸缓冲盐(PBS),其他各组均采用一次性注射干酪乳杆菌细胞壁提取物,建立KD小鼠模型.造模成功后,阿司匹林组,GL Ps低、高剂量组分别给予设定剂量的药物灌胃;对照组和KD模型组给予相应量的PBS,连续给药15 d.实验期间观察小鼠变化,实验结束后分离小鼠脾脏检测T淋巴细胞转化的刺激指数;流式细胞术检测T淋巴细胞亚群(CD_(3)^(+)、CD_(4)^(+)、CD_(4)^(+)/CD_(8)^(+))的含量;ELISA法检测小鼠外周血肿瘤坏死因子-α(TNF-α)、γ-干扰素(IFN-γ)、白细胞介素-2(IL-2)和白细胞介素-6(IL-6)等相关炎症因子的表达水平.结果给药结束后,与KD模型组比较,阿司匹林组、GLPs低、高剂量组小鼠体质量增加(P<0.05),体质量下降抑制率分别为11.817%、6.469%和7.203%;噻唑蓝(MTT)比色法检测结果显示GL Ps可抑制淋巴细胞的增殖转化功能,且高剂量组抑制效果明显优于低剂量组;同时,GLPs可以抑制淋巴细胞亚群CD_(4)^(+)、CD_(4)^(+)/CD_(8)^(+)的分化,上调CD_(3)^(+)含量,显著下调血清组织中TNF-α、IFN-γ、IL-2和IL-6的表达,并呈剂量依赖效应.结论灵芝多糖能够抑制淋巴细胞的异常活化,降低外周血组织中的炎症因子,能够双向调节川崎病小鼠机体免疫反应,且疗效与使用剂量有关,本研究可为川崎病的治疗提供相应的理论基础.OBJECTIVE To explore the effects of Ganoderma lucidum polysaccharides(GLPs)on regulation of T lymphocyte transformation and peripheral blood cytokines of mice models with Kawasaki disease(KD).METHODS The male BALB/c mice were randomly divided into the control group,the KD model group,the low-dose GLPs group(50 mg/kg),the high-dose GLPs group(100 mg/kg)and the aspirin group.The control group was injected with PBS,and other groups were injected with a one-time injection of Lactobacillus casei cell wall extract to establish the KD mice models.The aspirin group,the low-dose GLPs group and the high-dose GLPs group were given lavage with the specified doses of drug after the models were successfully built;the control group and the KD model group were given corresponding amounts of PBS for 15 consecutive days.The basic changes of the mice were observed during the experiment.After the experiment,the spleens of the mice were separated to detect stimulation indexes of T lymphocyte transformation,the contents of T lymphocyte subsets(CD_(3)^(+),CD_(4)^(+),CD_(4)^(+)/CD_(8)^(+))were determined by flow cytometry;the expression levels of related inflammatory factors in peripheral blood such as tumor necrosis factor-α(TNF-α),interferon-γ(IFN-γ),interleukin-2(IL-2)and IL-6 were detected by means of ELISA.RESULTS At the end of drug administration,the body weight of the aspirin group,the low-dose GLPs group and the high-dose GLPs group was significantly greater than that of the KD model group(P<0.05),the rates of inhibiting decrease of body weight were 11.817%,6.469%and 7.203%,respectively.The result of MTT test showed that GLPs could inhibit the proliferation and transformation of lymphocytes and that the inhibition effect of the high-dose GLPs group was better than that of the low-dose GLPs group;GLPs could inhibit the differentiation of lymphocyte subsets CD_(4)^(+)and CD_(4)^(+)/CD_(8)^(+),upregulate the content of CD_(3)^(+),and downregulate the expression of serum TNF-α,IFN-γ,IL-2 and IL-6 in a dose-dependent manner.CON
