摘  要:目的研究补体激活水平在脂多糖诱导急性乳腺炎小鼠乳腺中的变化。方法采用脂多糖诱导产后小鼠急性乳腺炎模型,观察不同感染时长模型组小鼠健康状况,血细胞分析法测定外周血各类炎症细胞数目,乳腺组织苏木精-伊红染色法(HE)染色,观察病理状况,酶联免疫法测定乳腺组织免疫球蛋白(IgG、IgA、IgM)表达水平、外周血补体蛋白(C3a、C4a、C5a)表达水平,Western blot检测Toll样受体(TLR1、TLR2)水平。结果模型组小鼠乳房出现水肿,乳汁分泌减少,抗拒哺乳;随着感染时间增加,外周血中炎症细胞数目呈升高趋势(P<0.05);病理切片观察结果显示,小鼠感染12 h后出现明显病理变化,腺泡壁变厚,出现炎症细胞浸润,外膜模糊粘连;乳腺组织免疫球蛋白(IgG、IgA、IgM)表达水平均呈升高趋势;外周血补体蛋白(C3a、C4a、C5a)表达水平总体为升高趋势,在感染12 h时,C3a、C5a有一个急剧上升的趋势;TLR1、TLR2受体与感染时间存在正相关。结论急性乳腺炎小鼠乳腺病理变化与乳腺组织中免疫球蛋白表达水平、补体激活水平、Toll样受体存在相关性,随着感染时间增加,乳腺组织被破环程度受补体激活水平影响。OBJECTIVE To observe the change of complement activation level in the mammary glands of mice with acute mastitis induced by lipopolysaccharide.METHODS The postpartum mice were induced by lipopolysaccharide to establish the acute mastitis models,the health status of the mice with different infection time was observed,the peripheral blood inflammatory cells were determined by using blood cell analysis method,the pathological conditions were observed by means of breast tissue HE staining,the expression levels of immunoglobulins(IgG,IgA,IgM)in mammary tissues and peripheral blood complement proteins(C3 a,C4 a,C5 a)were detected with the use of enzyme-linked immunosorbent assay,and the levels of Toll-like receptors(TLR1,TLR2)were determined by using Western blot.RESULTS In the model group,the breasts showed edema,the milk secretion decreased,and they resisted breastfeeding.With the increase of the infection time,the number of inflammatory cells in the peripheral blood was increased(P<0.05).The pathological section showed that the pathological changes were obvious after the mice were infected for 12 hours,with acinar walls thickened,inflammatory cells infiltrated,the outer membrane fuzzy and adhered.The expression levels of immunoglobulins(IgG,IgA,IgM)in breast tissues showed upward trends;the expression levels of peripheral blood complement protein(C3 a,C4 a,C5 a)showed upward trends,and the levels of C3 a and C5 a presented a sharply upward trend;the TLR1 and TLR2 receptors were positively correlated with the time of infection.CONCLUSION The pathological changes of mammary glands of the mice with acute mastitis are correlated with the levels of immunoglobulin expression,complement activation,and Toll-like receptors in the breast tissue.With the increase of infection time,the degree of disruption of mammary gland tissue is affected by the level of complement activation.
