RSV感染性毛细支气管炎与基质金属蛋白酶-3基因多态性的相关性分析

摘　　要：目的探讨基质金属蛋白酶-3(MMP-3)基因上rs522616位点及rs679620位点的多态性与呼吸道合胞病毒(RSV)感染性毛细支气管炎的易感性及严重性的相关性.方法纳入杭州汉族儿童269例,分为病例组(145例)和健康对照组(124例).病例组又分为轻、中度组(122例)和重度组(23例).所有儿童分为特异性组(54例)和非特异性组(215例).检测rs522616和rs679620位点的基因型,并进行基因型及等位基因频率分析.结果(1)在rs679620位点上,A等位基因在病例组出现的频率(36.9%)高于对照组(28.6%)(χ^(2)=4.127,P<0.05),AG基因型在重组出现的频率高于轻、中组(χ^(2)=7.931,P<0.05),在特异性体质组与非特异性体质组之间基因型及等位基因分布差异无统计学意义.(2)在rs522616位点上,病例组基因型频率及等位基因频率与对照组比较差异无统计学意义(P>0.05),且轻、中度组与重度组比较差异无统计学意义(P>0.05),但特异性组的AA基因型出现频率高于非特异性组(χ^(2)=8.679,P<0.05).结论杭州汉族儿童MMP-3基因rs679620位点的等位基因A可能与RSV感染性毛细支气管炎的易感性有关,而rs679620的基因型AG可能与疾病严重性有关;rs522616位点的基因多态性与RSV感染性毛细支气管炎的易感性及严重性均无明显关联,但基因型AA可能与特异性体质有关.Objective To explore the asocation of MMP-3 rs522616 and rs679620 polymophisms with the suscepibility and severity of RSV infection in Hangzhou.Methods 145 children with RSV infection between 2-24 months as case group.And 124 RSV negative healthy children as the control group.All children were divided into alrgic constitution group(54)and non-alrgic constution group(215).Case group was also divided into mild group(122)and sever group(23).Results 1.Rs679620:the A-alele frequency in case group was higher than control group(χ^(2)=4.127,P<0.05).And AG-genotype frequency in sever group was higher than mild group(χ^(2)=7.931,P<0.05).There were no significant differences of genotype frequencies between allergic constitution group and non-allergic constitution group(P>0.05).2.Rs522616:There were no significant differences of genotype frequencies between case group and control group(P>0.05)or between mild group and sever group(P>0.05).The frequency of AA-genotype in alrgic constitution group was higher than non-alrgic constitution group(χ^(2)=8.679,P<0.05).Conclusions A-alele on rs679620 on MMP-3 gene is related to the susceptibility of capillary bronchitis with RSV infection in children in Hangzhou.And AG-genotype on rs679620 on MMP-3 gene is related to the severity of capillary bronchitis with RSV infection.Gene polymorphism of rs522616 is not related to the suscepibility and severity of capillary bronchitis with RSV infection.But AA-genotype on rs522616 is related to the allergic constitution.
