摘  要:目的:探讨柴胡加龙骨牡蛎汤对七氟醚致老年动物认知损害的保护作用机制。方法:96只老年大鼠分成单纯七氟醚组(S组)、柴胡加龙骨牡蛎汤预处理组(B组)、柴胡加龙骨牡蛎汤预处理对照组(BC组)以及空白对照组(C组),每组24只。Morris水迷宫统计大鼠逃避潜伏期和跨台次数;TUNEL法对海马CA1区神经元计数;ELISA检测IL-1β、IL-6和TNF-α表达;Western blotting检测NF-κB p65表达。结果:1~4 d时,S组逃避潜伏期比C组长,B组逃避潜伏期则比S组短(P<0.05);S组跨台次数比C组少,B组跨台次数比S组多(P<0.05);S组神经元数量比C组少,B组神经元数量比S组多(P<0.05);S组IL-1β、IL-6和TNF-α表达比C组高,B组IL-1β、IL-6和TNF-α表达比S组低(P<0.05);S组NF-κB p65表达比C组高,B组NF-κB p65表达比S组低(P<0.05)。B组和BC组各指标均差异无统计学意义(P>0.05)。结论:柴胡加龙骨牡蛎汤对七氟醚致老年动物认知损害的保护可能是通过阻碍NF-κB通路的激活,降低炎症因子表达,减少神经元细胞凋亡来实现的。Objective:To investigate the protective mechanism of Chaihu Jia Longgu Muli decoction on sevoflurane induced cognitive impairment in aged animals.Methods:96 aged rats were divided into sevoflurane group(group S),Chaihu Jia Longgu Muli decoction pretreatment group(group B),control group of Chaihu Jia Longgu Muli decoction pretreatment group(group BC)and blank control group(group C),with 24 rats in each group.The escape latency and cross-stage times of rats were measured by Morris water maze.Neurons in hippocampal CA1 region were counted by TUNEL method.The expression of IL-1β,IL-6 and TNF-αwere detected by ELISA.The expression of NF-κB p65 was detected by Western blotting.Results:From 1 to 4 days,the escape latency of group S was longer than that of group C,while that of group B was shorter than that of group S(P<0.05);the number of crossover in group S was less than that in group C,and the number of crossover in group B was more than that in group S(P<0.05);the number of neurons in group S was less than that in group C,and the number of neurons in group B was more than that in group S(P<0.05);the expression of IL-1β,IL-6 and TNF-αin group S was higher than that in group C,and the expression of IL-1β,IL-6 and TNF-αin group B was lower than that in group S(P<0.05);the expression of NF-κB p65 in group S was higher than that in group C,and that in group B was lower than that in group S(P<0.05).There was no difference in each index between group B and BC(P>0.05).Conclusion:The protective effect of Chaihu Jia Longgu Muli decoction on sevoflurane-induced cognitive impairment in aged animals may be achieved by blocking the activation of NF-κB pathway,reducing the expression of inflammatory factors and reducing neuronal apoptosis.
