摘 要:为明确凤仙花授粉结实规律以及品系间的遗传关系,本研究利用单株自交、群体自交与开放杂交的方法验证凤仙花的结实习性为常异花授粉植物,心室数目变化在5~7的范围内,雄蕊的散粉时间集中在1 d内的14:00-15:00。通过ISSR-PCR分子标记对9种主栽凤仙花品系进行了多样性研究,利用11条ISSR引物对9种凤仙花DNA进行扩增,共得到77个位点,其中63个位点具有多态性,占到全部位点的81.81%。检测等位基因数(Na)平均为1.8261;有效等位基因数(Ne)在1.0000~1.9935之间,平均为1.4324;Nei’s基因多样性指数(H)介于0.1078~0.4984之间,平均为0.2617;Shannon’s信息指数(I)在0.2192~0.6915之间,平均为0.4005,证明凤仙花遗传多样性丰富。通过聚类分析发现,在阈值为0.58的地方,2号品系被单独分开,在阈值为0.65的地方,剩余8个品系被分为A和B两个类群。本研究针对凤仙花授粉结实的调查和多样性的研究可为日后获得株型、花色更加优良的凤仙花品种提供一定的理论依据。In order to clarify the pollination and fruiting law of Impatiens and the genetic relationship between the strain.In this study,used the methods of single plant self pollination,group self pollination and open hybridization to verify that the fruiting habit of Impatiens is an often cross pollinated plant,the number of ventricles changes in the range of 5~7,the time of stamen dispersal is from 14:00 to 15:00 in a day.The diversity of 9 major Impatiens strains was studied by ISSR-PCR,DNA amplification of 9 species of Impatiens by 11 ISSR primers,a total of 77 loci were obtained of which 63 loci were polymorphic,accounting for 81.81%of all loci.The average number of detected alleles(Na)was 1.8261,the average number of effective alleles(Ne)was 1.0000~1.9935,the average was1.4324,the Nei’s gene diversity index(H)was 0.1078~0.4984,the average was 0.2617,the Shannon’s information index(I)was 0.2192~0.6915,the average was 0.4005,indicating that the genetic diversity of Impatiens was rich.Through cluster analysis,we found that at the threshold of 0.58,line 2 was separated separately,and at the threshold of 0.65,the remaining 8 lines were divided into two groups A and B.The investigation and diversity of pollination and seed setting of Impatiens can provide a theoretical basis for obtaining better plant type and flower color in the future.
