摘 要:目的通过检测触觉阻断对小鼠恐惧建立和消退过程中僵直反应的影响,探讨交互模式可塑性与恐惧情绪的认知和调节之间的关系。方法选择出生后1 w的ICR小鼠,随机分为对照组和建模组。对建模组小鼠每日修剪胡须并维持至0.5 mm以阻断触觉,24 d后通过声音条件性和场景条件性恐惧建立和消退程序分别检测两组小鼠的僵直百分比,同时检测了触觉阻断对小鼠声音和场景条件性非特异性僵直行为以及自发活动的影响。结果声音条件性恐惧建立阶段,两组小鼠僵直百分比呈逐渐升高趋势,而建模组小鼠僵直百分比明显高于对照组(P<0.05);声音条件性恐惧消退和消退再现阶段,两组小鼠僵直百分比呈逐渐降低趋势,且建模组小鼠僵直百分比明显高于对照组(P<0.05);场景条件性恐惧建立阶段,两组小鼠僵直百分比呈逐渐升高趋势,但两组间僵直百分比没有差异;场景条件性恐惧消退和消退再现阶段,两组小鼠僵直百分比呈逐渐降低趋势,而建模组小鼠僵直百分比明显高于对照组(P<0.05);建模组小鼠声音和场景条件性非特异性僵直水平与对照组相比无显著性差异,且触觉阻断不影响小鼠自发活动。结论触觉阻断可影响小鼠恐惧情绪的建立和消退过程,其机制可能与交互模式可塑性引起的大脑皮层交互和重组改变了恐惧情绪的学习和记忆神经回路有关。Objective The purpose of this study was to investigate the effectsof whiskerdeprivation on fear establishment and extinction in mice,and to elucidate the mechanisms underlying cross-modal plasticity and fear acknowledge and regulation.Methods ICR mice(1 week old)were randomly divided into control group and model group.The whiskers of mice were clipped once in 24 hours,and maintained at 0.5mm length to deprive whisker tactile input.24 days latter,the percentage of stiffness in mice in both groups were detected through the establishment and disappear of auditory and contextualfear.We also detected the influence of whisker deprivation on auditory and contextual none specific stiffness behavior and spontaneous activity.Results Duringthe establishment phase,percentage of stiffness in mice in both groups presented increase tendency,and mice in deprivation group showed significantly higher stiffnesspercentage duringauditory fear establishment than the control group(P<0.05).The deprivation group also presented significantly higher percent of freezing during extinction and reappear phaseof auditory fear,and the percentage of stiffness in mice in both groups presented decrease trend(P<0.05).Duringestablishment phase of contextual fear,there was no difference in percent of freezing between two groups.Compared with the control group,miceofdeprivation group showed significantly higher percent of freezing during contextual fear extinction and reappear phase,and the percentage of stiffness in mice in both groups presented decrease trend(P<0.05).Whisker deprivation had no significant effects on nonspecific responses and spontaneous activity of mice in model group.Conclusions These results suggest that whisker deprivation could affect the process of fear establishment and extinction in mice,and the mechanism may be related to cortical interaction and reorganization induced by the plasticity of interaction patterns that alter the neural circuitry of learning and memory related to fear.
