摘  要:目的研究制备人嗜中性粒细胞肽1(human neutrophil peptide,HNP-1)脂质体,评价对细菌性肺炎的治疗作用,为耐药菌感染的细菌性肺炎类疾病的治疗提供新思路。方法采用逆向蒸发法制备HNP-1脂质体(Lipo-H),用于肺吸入制剂,通过透射电镜和马尔文粒度仪评价制备的Lipo-H。进一步考察体外对耐药菌的抗菌效果,建立大鼠细菌性肺炎模型并用于药效评价。结果将制成脂质体后,粒径为106.5nm,粒度分布均一,Zeta 电势为9.2mV,适合吸入给药。体外对细菌有明显抗菌效果,体内对细菌性肺炎有明显治愈作用。结论制成脂质体后,稳定性显著提升,同时通过肺吸入给药,增加药物的靶向性,药效明显改善,是细菌性肺炎治疗的一种新方法。Objective To prepare the liposomal human neutrophil peptide 1,evaluate its therapeutic effectson bacterial pneumonia,and provide a new idea for the treatment of bacterial pneumonia diseases infected by resistant bacteria.Methods Liposomal HNP-1(Lipo-H)were prepared by reverse evaporation method,which was used in lung inhalation preparations.The prepared Lipo-H was evaluated by transmission electron microscope and Malvern particle size analyzer.To investigate the antibacterial effect against drug-resistant bacteria in vitro,the rat model of bacterial pneumonia was established and used for drug efficacy evaluation.Results After being made into liposomes,the particle size was 106.5 nm,the particle size distribution was uniform,and the Zeta potential was 9.2 mV,which was suitable for inhalation administration.It has obvious antibacterial effect on bacteria in vitro,and has obvious cure effect on bacterial pneumonia in vivo.Conclusion After being made into liposomes,the stability is significantly improved,At the same time,it is administered through lung inhalation to increase the targeting of the drug,and the efficacy is significantly improved.It is a new method for the treatment of bacterial pneumonia.
