摘 要:目的探讨长链非编码RNA(LncRNA)在LADA和T2DM中的表达,并预测LncRNA作用的靶向基因,为LADA和T2DM的诊断及鉴别诊断提供基因表达谱。方法选取2017年7月至2018年6月于吉林大学第二医院内分泌科就诊的DM患者或正常糖耐量(NGT)人群,分为T2DM组、LADA组及NGT组,每组各4例。采用高通量测序检测,挑选表达差异有统计学意义的LncRNA构建基因表达谱。结果与NGT组比较,T2DM组LncRNA 68763个表达上调,28523个表达下调;LADA组LncRNA 68748个表达上调,28538个表达下调。各组LncRNA表达差异显著。在有统计学意义的数据中选取2个差异倍数最高的LncRNA(LncRNA ENST00000602845‐NCBP2、LncRNA ENST00000364558‐HECTD4)进一步分析,并通过GO、KEGG通路分析预测LncRNA的靶基因功能。结论LADA、T2DM患者LncRNA表达谱与NGT人群存在明显差异,可能通过多种机制参与发病。Objective To evaluate the LncRNA expression in type 2 diabetes mellitus(T2DM)and latent autoimmune diabetes in adults(LADA)and to predict the LncRNA target genes,in order to generate their expression profiles for the diagnosis as well as differential diagnosis of T2DM and LADA.Methods DM patients and normal glucose tolerance(NGT)subjects in Endocrinology Department in The Second Hospital of Jilin University from July 2017 to June 2018 were enrolled in this study.All the participants were divided into three groups:T2DM group(n=4),LADA group(n=4)and NGT group(n=4).LncRNA with significant differential expression was detected by high‐throughput sequencing and then selected to construct LncRNA gene expression profiles.Results A total of 68763 LncRNA were up‐regulated and 28523 were down‐regulated in T2DM group compared with NGT group.A total of 68748 LncRNAs were up‐regulated and 28538 were down‐regulated in LADA group compared with NGT group.The expression of LncRNAs was significantly different among the three groups.Two significantly changed LncRNAs were obtained(LncRNA ENST00000602845‐NCBP2 and LncRNA ENST00000364558‐HECTD4)and the target gene of LncRNA was predicted by gene ontology(GO)analysis and pathway analysis in Kyoto Encyclopedia of Genes and Genomes(KEGG).Conclusion LncRNA expression profiles were significantly different in LADA and T2DM patients compared with NGT subjects,and may be involved in the pathogenesis of diabetes through multiple mechanisms.
