摘 要:目的:建立短柄五加根、茎中绿原酸的含量测定方法,比较不同产地短柄五加中绿原酸的含量。方法:采用Agilent TC-C18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.1%磷酸溶液,梯度洗脱;流速为1.0 mL/min;柱温30℃;检测波长348 nm;进样体积10μL。结果:绿原酸在5.81~290.25μg/mL范围内呈良好的线性关系,R2=0.9992,短柄五加根、茎中绿原酸平均回收率为98.75%,98.05%。短柄五加根、茎中绿原酸含量分别为陕西宜君县(1.914 mg·g^(-1)、2.322 mg·g^(-1)),甘肃正宁县(2.363 mg·g^(-1)、2.442 mg·g^(-1)),陕西黄龙县(3.769 mg·g^(-1)、2.642 mg·g^(-1)),陕西旬邑县(1.903mg·g^(-1)、2.723mg·g^(-1))。结论:该方法快速、准确,测定结果显示黄龙县短柄五加根中绿原酸含量最高,整体比较根、茎中绿原酸含量无显著差异,基本都在2.5 mg·g^(-1)左右。Objective:To establish a method for the determination of chlorogenic acid in root and stem of Acanthopanax brachypus Harms and compare the content of Chlorogenic acid in Acanthopanax brachypus Harms from different habitats.Methods:Seperation was carried out on Agilent TC-C18 column(250 mm×4.6 mm,5μm);The mobile phase concisted of acetonitrile-0.1%phosphoric acid solution with gradient elution;The flow rate was 1.0 mL/min,temperature of column was 30℃,the detection wavelength were set at 348 nm,and volume of injection was 10μL.Results:The linear range of chlorogenic acid was 5.81~290.25μg/mL,R2=0.9992.The average recovery of Chlorogenic acid in root and stem of Acanthopanax brachypus Harms were 98.75%and 98.05%.The contents of Chlorogenic acid in root and stem of Acanthopanax brachypus Harms were 1.914 mg·g^(-1)and 2.322 mg·g^(-1)in Yijun County,2.363 mg·g^(-1)and 2.442 mg·g^(-1)in Zhengning County,3.769 mg·g^(-1)and 2.642 mg·g^(-1)in Huanglong County,1.903 mg·g^(-1)and 2.723 mg·g^(-1)in Xunyi County.Conclusion:The method is rapid and accurate.The content of Chlorogenic acid in the root of Acanthopanax brachypus Harms in Huanglong county is the highest,and there is no significant difference in the content of Chlorogenic acid in the root and stem,which is about 2.5 mg·g^(-1).
