摘　　要：目的研究探讨lnc RNA XIST在胶质瘤中的表达水平及其生物学功能。方法选取2018年6月—2019年5月的35例胶质瘤患者为观察组,同时期的35例脑外伤患者为对照组。比较两组、观察组中不同分级者的组织lnc RNA XIST表达情况,同时检测转染siRNA XIST与siRNA-NC的U87及U251细胞克隆数、凋亡细胞比率及细胞穿膜数。结果观察组的lnc RNA XIST表达高于对照组,观察组中不同分级者的组织lnc RNA XIST表达比较,差异具有统计学意义(P<0.05),siRNA XIST的U87及U251细胞克隆数及细胞穿膜数低于siRNA-NC,凋亡细胞比率高于siRNA-NC,差异具有统计学意义(P<0.05)。结论lnc RNA XIST在胶质瘤中的表达水平较高,lnc RNA XIST的高表达状态提升了胶质瘤细胞的增殖及侵袭能力。Objective To study and investigate the expression levels of lnc RNA XIST in glioma and its biological function.Methods 35 patients with glioma from June 2018 to May 2019 were chosen as the observation group,35 patients of same ages with traumatic brain injury were chosen as the control group.Then the tissue lnc RNA XIST in glioma expression situation of two groups and observation group with different classifications were compared,and the U87 and U251 cell clones,apoptotic cells ratio and number of cell penetrating membrane of transfection siRNA AGAP2-AS1 and siRNA-NC were compared.Results The tissue lnc RNA XIST expression of observation group was significantly higher than that of control group,the tissue lnc RNA XIST expression of observation group with different classifications were compared,there were statistically significant differences(P<0.05),the U87 and U251 cell clones and number of cell penetrating membrane of transfection siRNA AGAP2-AS1 were significantly lower than those of siRNA-NC,the apoptotic cells ratio was significantly higher than that of siRNA-NC,there were statistically significant differences(P<0.05).Conclusion The expression levels of lnc RNA XIST in glioma and its biological function is higher,and the high expression of lnc RNA XIST enhances the proliferation and invasion of glioma cells.
