摘  要:利用MTT试验,检测石蒜碱对人肝癌HepG-2细胞具有增殖抑制作用,并观察细胞凋亡形态,采用流式细胞仪检测肿瘤细胞凋亡率,发现石蒜碱对人肝癌HepG-2肿瘤细胞具有显著的抗肿瘤作用;通过对肿瘤细胞膜上主要组成成分进行检测,发现石蒜碱可以改变HepG-2细胞膜主要组成成分,使细胞膜结构发生变化;通过考察石蒜碱对HepG-2细胞细胞膜结构功能的影响,发现石蒜碱可以破坏肿瘤细胞膜完整性,并且能够降低肿瘤细胞膜封闭度、肿瘤细胞膜流动性以及肿瘤细胞膜阳离子通道活性,肿瘤细胞膜主要组成成分、结构的改变将会改变肿瘤细胞的内环境,细胞的内环境的改变必将影响细胞正常的新陈代谢功能,引发细胞凋亡.This paper used the MTT test to detect the proliferation inhibitory effect of lycorine on human liver cancer HepG-2 cells,and observe the morphology of cell apoptosis.Flow cytometry was used to detect the apoptosis rate of tumor cells,and it was found that lycorine was harmful to humans.Liver cancer HepG-2 tumor cells have a significant anti-tumor effect;and through the detection of the main components of the tumor cell membrane,it was found that lycorine can change the main components of the HepG-2 cell membrane and change the cell membrane structure;the effect on the structure and function of the cell membrane of HepG-2 cells.It was found that lycorine can destroy the integrity of tumor cell membranes,and can reduce tumor cell membrane sealing,tumor cell membrane fluidity and tumor cell membrane cation channel activity,and changes in the main components and structure of tumor cell membranes It will change the internal environment of tumor cells.Changes in the internal environment of cells will definitely affect the normal metabolic function of cells,and these changes will trigger cell apoptosis.
