摘　　要：采用酶法提取作为前处理,高效液相色谱-原子荧光光谱分析新疆(南疆)冻干驴乳粉中硒形态含量。酶法提取最佳条件为温度40℃、时间3 h、酶添加量STD3((蛋白酶、脂肪酶、淀粉酶均0.06 g)、pH为7,流动相为5 mmoL柠檬酸+0.5 mmoL TBAB+2%CH3OH溶液作为流动相(pH=4.5)。此方法硒代蛋氨酸(SeCys)、亚硒酸根(Se(Ⅳ))、硒代胱氨酸(SeMet)、硒酸根(Se(Ⅵ))实现了基线分离,方法检出限在0.12~1.00μg/kg之间、RSD在0.98%~1.89%之间、样品空白加标回收率在样品加标回收率在30.5%~95.2%。结果显示,新疆南疆冻干驴乳粉中硒含量丰富,有机硒含量在16.68~18.60μg/kg之间,占总硒含量的94.4%~100.0%,含有很少或者不含无机硒。The content of selenium in freeze-dried donkey milk powder was determined by HPLC-ICP-MS.The optimum conditions of enzymatic extraction were temperature 40℃,time 3 h,enzyme addition amount(0.06 g for protease,lipase and amylase),pH=7,mobile phase 5 mmol citric acid+0.5 mmol TBAB+2%CH3OH solution as mobile phase(pH=4.5).SeCys,Se(Ⅳ),SeMet,Se(Ⅵ)were separat⁃ed by baseline.The detection limit of the method was 0.12μg/kg~1.00μg/kg,RSD was 0.98%~1.89%,and the recovery of blank standard addition was 30.5%~95.2%.The results showed that the content of selenium in the freeze-dried donkey milk powder in southern Xinjiang was rich and beneficial to human body.The content of organic selenium ranged from 16.68~18.60μg/kg,accounting for 94.4%~100.0%of the total selenium content,containing little or no inorganic selenium.
