甘露消毒丹抗甲型H1N1流感病毒感染体内外作用评价

摘　　要：目的:观察甘露消毒丹抗甲型H1N1流感病毒感染体内外作用,为流行性感冒治疗提供实验依据。方法:体外实验,以甲型H1N1流感病毒鼠肺适应株UIXD-P18-2(UI182)致犬肾细胞(MDCK)病变,采用ReedMuench法计算TCID_(50),采用MTT法测定甘露消毒丹对MDCK的TC_(0)和TC_(50),采用Probit回归分析计算甘露消毒丹及达菲对病毒感染的EC_(50)与TI;体内实验,以UI182滴鼻感染小鼠,从攻毒起14d,观察并计算小鼠死亡保护率、生命延长率、肺指数,检测其肺组织病毒滴度。结果:UI182的TCID_(50)为10^(-2.82)/0.1mL,甘露消毒丹对MDCK的TC_(50)为370.8μg/mL,TC_(0)为15.63μg/mL。甘露消毒丹预防组、甘露消毒丹抗病毒吸附组、甘露消毒丹治疗组以及达菲治疗组的EC_(50)分别为(3.89±0.55)、(2.86±0.20)、(2.44±0.18)、(2.04±0.23)μg/mL,TI值分别为95.32、129.65、151.96及181.76。甘露消毒丹能提高小鼠死亡保护率,抑制肺损伤,降低肺指数和肺组织病毒滴度。结论:甘露消毒丹具有体内外抗甲型H1N1流感病毒作用。Objective:To observe the effect of Ganlu Xiaodu Pill against influenza A(H1N1)virus in vitro and in vivo and provide experimental basis for influenza treatment.Methods:In-body test,MDCK was infected by mouse lung adapt strain UI182 of influenza A(H1N1)virus.TCID_(50) after infection was calculated by Reed-Muench method.TC_(0) and TC_(50) of mannan disinfection pills for MDCK were measured by MTT colorimetry.EC_(50) and TI of mannan disinfection pills and Tamiflu for virus infection were calculated by Probit regression analysis.Bit regression analysis calculated EC_(50) and TI for viral infections of Ganlu Xiaodu Pill and Duffy.In vivo tests of mice,the death protection rate,life extension rate,lung index and pulmonary virus titration were observed,calculated and determined after challenge with UI182 nasal infection in 14 days.Results:The TCID_(50) of UI182 was 10^(-2.82)/0.1 mL,the TC_(50) of MDCK was 370.8μg/mL,TC_(0) was 15.63μg/mL.The EC_(50) of Ganlu Xiaodu Pill prevention group,antiviral adsorption group,treatment group and oseltamivir treatment group were(3.89±0.55),(2.86±0.20),(2.44±0.18),and(2.04±0.23)μg/mL,respectively,and the TI values were 95.32,129.65,151.96 and 181.76,respectively.Ganlu Xiaodu Pill could increase the mortality protection rate of mice infected,inhibit lung damage,and reduce the level of virus in the lungs of mice.Conclusion:Ganlu Xiaodu Pill has obvious in vivo and in vitro against-influenza A(H1N1)virus effect.
