摘 要:目的:研究健脾化浊消脂方防治高脂饲料诱导的大鼠非酒精性脂肪性肝炎(NASH)的作用及其机制。方法:32只雄性SD大鼠随机分为3组:正常对照组(8只)、健脾化浊消脂方组(8只)、模型对照组(16只)。正常对照组给予常规普通饲料喂养,其余两组采用高脂饲料喂养复制NASH大鼠模型。于第5周,健脾化浊消脂方组予健脾化浊消脂方12.5g·kg^(-1)·d^(-1)灌胃,其余两组予以等体积的0.9%氯化钠灌胃,共干预12周。末次给药后24h,检测大鼠血清甘油三酯(TG)、游离脂肪酸(FFA)含量;取肝脏组织行HE、油红O染色评价病理改变;鲎试剂法检测血浆脂多糖(LPS)含量;ELISA法检测血清白介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)的水平。结果:与正常对照组比较,模型对照组大鼠体质量和肝脏指数显著上升(P<0.01),血清TG、FAA显著升高(P<0.01),肝组织NAS加重(P<0.01),血浆LPS和血清TNF-α、IL-1β含量显著上升(P<0.01)。与模型对照组比较,健脾化浊消脂方组大鼠体质量和肝脏指数显著降低(P<0.05,P<0.01);血清TG、FAA水平显著下降(P<0.05,P<0.01);肝组织NAS积分显著改善(P<0.01);LPS和TNF-α、IL-1β含量显著降低(P<0.05,P<0.01)。结论:健脾化浊消脂方可有效阻止NASH进展,通过抑制肠源性内毒素血症,缓解慢性代谢性炎症可能是其防治NASH的作用机制之一。Objective:To investigate the effects and mechanism of Jianpi Huazhuo Xiaozhi Formula(JPHZXZ)on the prevention and treatment of non-alcoholic steatohepatitis(NASH)in rats induced by high-fat diet.Methods:Thirty-two male SD rats were randomly divided into 3 groups:normal control group(8 rats),JPHZXZ group(8 rats)and model control group(16 rats).The normal control group was fed with conventional diet,and the other two groups were fed with high fat diet to reproduce the NASH model.In the 5 th week of the experiment,the JPHZXZ group was given JPHZXZ at a dose of 12.5 g·kg^(-1)·d^(-1) by gavage,the equal volume of 0.9%sodium chloride of equal volume was given to the other 2 groups for 12 weeks.24 h after last administration,serum triglyceride(TG)and free fatty acid(FFA)were measured;Liver tissues were evaluated for pathological changes by HE and oil red O staining to evaluate the pathological changes;Plasma lipopolysaccharide(LPS)was measured by limulus reagent method;Interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)were measured by ELISA.Results:Compared with the normal control group,the body weight and liver index of the model control group were increased(P<0.01);The serum TG and FAA of the model control group were raised(P<0.01);The NAS score of the liver tissue was worsen(P<0.01);The plasma levels of LPS and serum TNF-αand IL-1βwere also increased(P<0.01).Compared with the model control group,the body mass and liver index of the rats in the JPHZXZ group were significantly lowered(P<0.05,P<0.01);The serum TG and FAA levels were also significantly decreased(P<0.05,P<0.01);NAS scores were significantly improved(P<0.01);LPS and TNF-α,IL-1βlevels were significantly reduced(P<0.05,P<0.01).Conclusion:JPHZXZ could effectively prevent the development of NASH,inhibiting enterogenous endotoxemia and relieving chronic metabolic inflammation may be one of the mechanisms of its prevention and treatment of NASH.
