摘 要:目的:研究益气温阳护卫汤对哮喘大鼠Th17/Treg细胞失衡与RORγ、Foxp3的影响。方法:将144只SPF级雄性SD大鼠随机分为对照组,模型组,益气温阳护卫汤低、中、高剂量组及地塞米松组,卵蛋白氢氧化铝溶液致敏,雾化吸入卵蛋白激发建立哮喘大鼠模型,分别于激发1、2、4周末各组处死大鼠8只,检测肺组织中Th17、Treg细胞比例及RORγ、Foxp3 mRNA及蛋白表达。结果:与对照组同期比较,模型组Th17细胞比例显著升高(P<0.01),Treg细胞比例显著降低(P<0.01);与同期模型组比较,中、高剂量组及地塞米松组Th17细胞比例均显著降低(P<0.01)、Treg细胞比例显著升高(P<0.05,P<0.01)。与同期对照组比较,模型组RORγmRNA及蛋白表达显著升高(P<0.05,P<0.01),Foxp3 mRNA及蛋白相对表达显著降低(P<0.05,P<0.01);与同期模型组比较,中、高剂量组及地塞米松组RORγmRNA及蛋白相对表达量均显著降低(P<0.05,P<0.01),Foxp3mRNA及蛋白相对表达量均显著升高(P<0.05,P<0.01)。结论:益气温阳护卫汤能有效改善哮喘大鼠Th17/Treg细胞失衡,其机制可能与调节特异性转录因子RORγ、Foxp3的表达相关。Objective:To study the effects of Yiqi Wenyang Huwei Decoction on Th17/Treg cell imbalance,RORγand Foxp3 in asthmatic rats.Methods:A total of 144 SPF male SD rats were randomly divided into control group,model group,and low,medium,and high dose group of Yiqi Wenyang Huwei Decoction,and dexamethasone group.The ovalbumin aluminum hydroxide solution was sensitized and sprayed inhaled.Aerosol inhalation of ovalbumin was used to establish the asthma rat model.Eight rats in each group were sacrificed at the end of one week,two weeks and four weeks after the challenge.The proportion of Th17 and Treg cells in lung tissues and the expression of RORγand Foxp3 mRNA and protein were detected.Results:Compared with the control group,the proportion of Th17 cells in the model group was significantly increased in the same period(P<0.01),and the proportion of Treg cells was significantly decreased(P<0.01);Compared with the model group,the proportion of Th17 cells in the medium,high-dose group and the dexamethasone group was significantly reduced(P<0.01),and the proportion of Treg cells was significantly increased(P<0.05,P<0.01).Compared with the control group,the expression of RORγmRNA and protein in the model group at the same stage was significantly increased(P<0.05,P<0.01),and the relative expression of Foxp3 mRNA and protein was significantly reduced(P<0.05,P<0.01).Compared with the model group,the relative expressions of RORγmRNA and protein in the middle and high-dose groups and the dexamethasone group were significantly reduced(P<0.05,P<0.01),and the relative expressions of Foxp3 mRNA and protein were significantly increased(P<0.05,P<0.01).Conclusion:Yiqi Wenyang Huwei Decoction can effectively improve the Th17/Treg cell imbalance in asthmatic rats,and its mechanism may be related to the regulation of the expression of specific transcription factors RORγand Foxp3.
