隔药灸脐法对腹泻型肠易激综合征患者micro RNA的表达调控作用

摘　　要：目的:探究隔药灸脐法对腹泻型肠易激综合征(IBS-D)患者microRNA(miRNA)的调控作用。方法:以纳入的20例IBS-D患者为隔药灸脐组,以年龄、性别相匹配的10名健康志愿者为正常对照组。IBS-D患者给予隔药灸脐法治疗,每周1次,共8周。健康志愿者不给予任何治疗。检测隔药灸脐组治疗前与正常对照组,以及隔药灸脐组治疗前后外周血中miRNA的差异表达谱,并利用RT-PCR法验证。结果:芯片分析显示隔药灸脐组治疗前与正常对照组相比,有5个miRNAs表达差异显著,其中4个下调,1个上调;隔药灸脐组治疗后与治疗前相比,有5个miRNAs表达差异显著且均为上调。RT-PCR结果显示隔药灸脐组治疗后较治疗前可上调hsa-let-7b-5p、hsa-miR-19b-3p、hsa-miR-20a-5p的表达(P<0.05)。结论:隔药灸脐法可能通过上调hsa-let-7b-5p、hsa-miR-19b-3p、hsamiR-20a-5p的表达而达到治疗效果,为隔药灸脐法治疗IBS-D提供了基因表达调控方面的证据。Objective:To explore the regulation of microRNA(miRNA)expression in IBS-D patients treated with herb-partitioned moxibustion on the navel.Methods:The 20 patients diagnosed as IBS-D were assigned to the herbpartitioned moxibustion on the navel group and treated with herb-partitioned moxibustion on the navel,once a week,with 8 weeks,and 10 healthy volunteers matched in age and gender were assigned to the normal control group were not given any treatment.The differential expression profiling of miRNA in peripheral blood of IBS-D patients and healthy volunteers were detected,and RT-PCR was used for verification.Results:Microarray results showed that there were significant differences in the expression of 5 miRNAs in peripheral blood of IBS-D patients before treatment compared with the normal control group.After treatment,there were significant differences in the expression of 5 miRNAs which were all upregulated in peripheral blood of IBS-D patient.RT-PCR results showed that the expressions of hsa-let-7 b-5 p,hsa-miR-19 b-3 p and hsa-miR-20 a-5 p could be up-regulated after the treatment of the herb-partitioned moxibustion on the navel(P<0.05).Conclusion:Herb-partitioned moxibustion on the navel may achieve therapeutic effects by up-regulating the expression of hsa-let-7 b-5 p,hsa-miR-19 b-3 p and hsa-miR-20 a-5 p,providing evidence for the regulation of gene expression in the treatment of IBS-D.
