摘  要:目的:建立HPLC-ELSD同时测定闹羊花中闹羊花毒素Ⅲ、闹羊花毒素Ⅱ、金丝桃苷、槲皮苷的含量测定方法。方法:采用HPLC-ELSD色谱法,Waters C18色谱柱(250mm×4.6mm,5μm);流动相为甲醇(A)和水(B)进行梯度洗脱,流速1.0mL/min,柱温35℃,ELSD参数:雾化温度40℃,漂移管温度85℃,气体压力25psi,气体流速1.5SLM。结果:4种成分线性回归方程的相关系数为0.9990~0.9996,平均回收率(n=9)为97.52%~100.41%,RSD为1.38%~2.22%。结论:该方法简便、准确、可靠,可为闹羊花的质量控制提供参考。Objective:To establish a quantitative method for determining the contents of rhodojaponinⅢ,rhodojaponinⅡ,hyperoside and quercitroside in Rhododendron molle G.Don.by HPLC-ELSD method.Methods:The HPLC-ELSD analysis was performed on Waters C18 column(250 mm×4.6 mm,5μm).The mobile phase was composed of methanol(A)and water(B),and gradient elution was carried out.The flow rate was 1.0 mL/min.The column temperature was maintained at 35℃.ELSD detector parameter:atomization temperature was 40℃.Drift tube temperature was 85℃.Gas pressure was 25 psi,and flow rate of carrier gas was 1.5 SLM.Results:The correlation coefficients of the four compounds ranged from 0.9990 to 0.9996.The average recovery rates(n=9)were between 97.52%and 100.41%,and the RSD were between 1.38%and 2.22%.Conclusion:The proposed method is simple,accurate and reliable,thus providing basis for comprehensive quality control of Rhododendron molle G.Don..
