摘 要:目的:用细胞学方法评价南葶苈子与白芥子的寒热药性。方法:用噻唑蓝(MTT)比色法考察南葶苈子与白芥子对HepG2细胞、SMMC7721细胞、HCT116细胞体外增殖的影响,倒置显微镜观察南葶苈子与白芥子对细胞形态的影响,台盼蓝染色法分析南葶苈子与白芥子的细胞毒作用。结果:南葶苈子在2~800μg/mL浓度范围内对3种细胞的生长增殖表现出抑制作用,随着浓度的增大,抑制率增大,判定为寒性;白芥子在2~800μg/mL浓度范围内对3种细胞的生长增殖表现出低浓度促进、高浓度抑制作用,判定为热性。南葶苈子能使细胞密度减小,固缩变圆;白芥子低浓度下使细胞密度增加,细胞结构致密,生长旺盛,高浓度下使细胞密度降低,固缩变圆。南葶苈子与白芥子对于这3种细胞没有细胞毒作用。结论:南葶苈子和白芥子寒热药性细胞学方法评价结果与文献记载一致;细胞学方法可用于南葶苈子与白芥子寒热药性研究。Objective:To evaluate the hot&cold characteristics of Descurainia sophia(L.)Webb ex Prantl and Sinapis alba L.by cellular models.Methods:MTT assay was used to investigate the effect of Descurainia sophia(L.)Webb ex Prantl and Sinapis alba L.on the proliferation of HepG2 cells,SMMC7721 cells and HCT116 cells in vitro.The morphological changes were observed through inverted microscope.Trypan-blue staining was used to analyze the cytotoxicity of Descurainia sophia(L.)Webb ex Prantl and Sinapis alba L.Results:Descurainia sophia(L.)Webb ex Prantl could inhibite the growth and proliferation of the cells in the concentration range of 2~800μg/mL,that means it possess cold characteristics;Sinapis alba L.could promote the growth and proliferation of the cells at low concentration,and inhibite the growth and proliferation at high concentration.The results means that Sinapis alba L.possess hot characteristics.Cells treated with Descurainia sophia(L.)Webb ex Prantl showing decreased the cell density.Sinapis alba L.increased the cell density at high concentration.Descurainia sophia(L.)Webb ex Prantl and Sinapis alba L.had no cytotoxic effect on the cells.Conclusion:The results of hot&cold characteristics of Descurainia sophia(L.)Webb ex Prantl and Sinapis alba L.obtained in this paper are consistent with traditional literature records.It demonstrated that the simple and reliable cytological method could be used to study the hot&cold characteristics of Descurainia sophia(L.)Webb ex Prantl and Sinapis alba L.
