摘  要:以L-脯氨酸和氯乙酰氯为原料合成维格列汀的重要中间体(S)-1-(2-氯乙酰基)吡咯烷-2-甲酸(1)。甲醇体系室温挥发14 d培养得到1的无色透明块状晶体,通过单晶X-射线衍射技术对其进行结构解析。结果表明1晶胞属于正交晶系,P2_(1)2_(1)2_(1)空间群,分子式为C_(7)H_(10)NO_(3)Cl,相对分子质量为191.61 g/mol,立体构型和预测构型一致,确证了1的空间结构。(S)-1-(2-chloroacetyl)pyrrolidine-2-carboxylic acid(1)is an important intermediate of vildagliptin,synthesized from L-proline and chloroacetyl chloride as raw materials.The title compound 1 colorless transparent massive crystal has been grown by evaporation in methanol for 14 d at room temperature.Then,the crystal structure was solved by single crystal X-ray diffraction(SCXRD)analysis.SCXRD confirmed the orthorhombic lattice with space group P2_(1)2_(1)2_(1) in 1 crystal.Moreover,the chemical formula and relative molecular mass were C_(7)H_(10)NO_(3)Cl and 191.61 g/mol,respectively.The measured crystal structure was in good agreement with the predicted one,confirming the single crystal structure of 1.
