龟鹿二仙膏对阳虚型少弱精子症大鼠的改善作用及CFTR蛋白表达的影响

摘　　要：目的:观察龟鹿二仙膏对少弱精子症大鼠的改善作用以及睾丸囊性纤维化跨膜传导调节因子(CFTR)蛋白表达的影响。方法:将50只雄性SD大鼠随机分为正常组、模型组、龟鹿二仙膏组、右归丸组和左卡尼汀组,除正常组外,其余均采用灌胃腺嘌呤造成少弱精子症模型,正常组和模型组给予生理盐水,其余各组灌胃相应药物28 d,最后一次灌胃后24 h处死,剪碎附睾,观察各组精子浓度及活力;采用Western Blot检测睾丸CFTR蛋白表达。结果:与模型组相比,龟鹿二仙膏组大鼠,阳虚症状得到相应的改善;并且龟鹿二仙膏组大鼠的睾丸组织得到一定程度的修复。正常组、龟鹿二仙膏组、右归丸组、左卡尼汀组精子密度、精子活力均高于模型组,差异具有统计学意义(P<0.05);正常组、龟鹿二仙膏组、右归丸组、左卡尼汀组的CFTR蛋白的表达量均高于模型组,差异具有统计学意义(P<0.05)。结论:龟鹿二仙膏能改善腺嘌呤所致少弱精子症大鼠的精子密度及活力,并且提高了其睾丸CFTR蛋白的表达。Objective:To observe the effect of Guilu Erxian Ointment on the improvement of oligoasthenospermia and the expression of testicular CFTR protein in the model rats.Methods:50 male SD rats were randomly divided into the normal group,the model group,the Guilu Erxian Ointment group,the Yougui Pill group and the Levocarnitine group.The oligoasthenospermia model was established by the administration of adenine.The normal group and the model group were given normal saline,and the remaining groups were treated with corresponding medications for 28 days.The rats were sacrificed 24 hours after the last administration,the epididymis was cut and the concentration and the vitality of sperm were observed.Western Blot method was used to test the expression of testicular CFTR protein.Results:The Yang deficiency symptoms were improved and the testis tissues were repaired to a certain degree in the Guilu Erxian Ointment group compared to those in the model group.The sperm density and sperm motility were significantly higher in the normal group,the Guilu Erxian Ointment group,the Yougui Pill group and the Levocarnitine group than those in the model group(P<0.05).The expression of CFTR protein was also obviously higher in the normal group,the Guilu Erxian Ointment group,the Yougui pill group and the L-carnitine group than that in the model group(P<0.05).Conclusion:Guilu Erxian Ointment can improve the sperm density and sperm motility in the rats with oligoasthenospermia caused by adenine,and it can increase the expression of testicular CFTR protein.
