摘  要:为了研究菌腔虫瘿中暗褐网柄牛肝菌(Phlebopus portentosus)菌丝体群体遗传结构,利用ITS、tef 1和rpb 1序列对云南、四川两省12个地理居群共45份样品进行系统发育、遗传多样性,地理居群遗传分化、变异分析。结果表明:从45份样品中,共检出15个ITS单倍型、16个tef 1单倍型和17个rpb 1单倍型,总体上单倍型多样性较高(Hd:0.7790~0.8460),核苷酸多样性较低(Pi:0.00178~0.00295)。地理居群DF、GD、HS、LC、MH、XMY和YB样品遗传分化程度高(F_(st)>0.25000),与其他地理居群样品缺乏基因交流(N_(m)<1.00),地理居群间的变异占总变异的69.73%,在系统发育树中按照地理来源聚在同一分支上;DZ、JN、YX、RZS样品遗传分化程度低(F_(st)<0.25000),相互间存在明显基因流(N_(m)>1.00),MY与DZ间也存在明显基因流(N_(m)=2.17);DZ、JN、YX、RZS与MY样品之间的变异仅占总变异的18.87%,在系统发育树中未严格按照地理来源聚类;地理居群样品间的遗传分化程度与地理距离无相关性(R^(2)=0.0081)。In order to study the population genetic structure of Phlebopus portentosus mycelia isolated from fungus-insect galls,ITS,tef 1 and rpb 1 sequences of 45 samples collected from 12 geographical populations in Yunnan and Sichuan provinces were analyzed for their phylogeny,genetic diversity,population genetic differentiation and genetic variation.The results showed that a total of 15 ITS haplotypes,16 tef 1 haplotypes and 17 rpb 1 haplotypes were detected in the samples,with a high haplotype diversity(Hd:0.7790~0.8460)and a low nucleotide diversity(Pi:0.00178~0.00295).Genetic differentiation index(F_(st))values of DF,GD,HS,LC,MH,XMY and YB between other geographical populations were high(F_(st)>0.25000),and their gene flow between other geographical populations were very limited(N_(m)<1.00).They were clustered within the same branch according to their geographical origins in the neighbor-joining phylogenetic tree.The molecular variation among DF,GD,HS,LC,MH,XMY and YB geographical populations accounted for 69.73%of the total variation.On the other hand,genetic differentiation among DZ,JN,YX and RZS were low(F_(st)<0.25000)with significant gene flow between each other(N_(m)>1.00).There was also gene flow between MY and DZ(N_(m)=2.17).The molecular variation among the remaining five populations DZ,JN,YX,RZS and MY accounted for 18.87%of the total variation,and they were not strictly clustered according to their geographical origins in the neighbor-joining phylogenetic tree.There was no correlation between genetic differentiation index among populations and geographical distance(R^(2)=0.0081).
