摘  要:目的探究miR-126抑制物对卵清蛋白诱导的哮喘小鼠免疫平衡和肺组织纤维化以及胶原沉积的影响.方法通过卵清蛋白(ovalbumin,OVA)诱导建立哮喘小鼠模型,转染miR-126 inhibitor-NC及miR-126 inhibitor至小鼠.HE染色检测肺组织病理损伤并进行肺组织炎症评分;检测支气管肺泡灌洗液(BALF)中炎性细胞数、总IgE及OVA特异性IgE含量;ELISA检测BALF中IL-12、IFN-γ、IL-4和IL-13水平;Masson染色检测肺纤维化;试剂盒检测HYP含量;Western印迹检测肺组织中α-SMA、Fibronectin、TGF-β1的表达.结果与Control组比较,OVA组肺组织肺损伤评分及炎症评分升高,白细胞总数、嗜酸性粒细胞、淋巴细胞和嗜中性粒细胞升高,总IgE及OVA特异性IgE含量升高,IL-12、IFN-γ、IL-4和IL-13水平升高,支气管上皮下胶原沉积增多,HYP含量升高,α-SMA、Fibronectin、TGF-β1表达升高,差异有统计学意义(P<0.05).与OVA组比较,OVA+miR-126 inhibitor组肺组织损伤减轻,肺损伤评分及炎症评分降低,白细胞总数、嗜酸性粒细胞、淋巴细胞和嗜中性粒细胞降低,总IgE及OVA特异性IgE含量降低,IL-12、IFN-γ水平降低,IL-4、IL-13水平升高,上皮下胶原沉积情况减轻,HYP含量降低,α-SMA、Fibronectin、TGF-β1表达降低,差异有统计学意义(P<0.05).结论miR-126抑制物改善卵清蛋白诱导的哮喘小鼠免疫平衡和肺组织纤维化以及胶原沉积.Objective To investigate the effects of miR-126 inhibitor on immune balance,pulmonary fibrosis and collagen deposition in ovoalbumin induced asthma mice.Methods Asthma mice were induced by ovalbumin.The mice were transfected with miR-126 inhibitor-NC and miR-126 inhibitor.HE staining was used to detect pathological injury of lung tissue and lung tissue inflammation score.The inflammatory cell count,total IgE and OVA-specific IgE content in bron-choalveolar lavage fluid(BALF)were measured.The levels of IL-12,IFN-γ,IL-4 and IL-13 in BALF were detected by ELISA.Masson staining was used to detect pulmonary fibrosis.The content of HYP was detected by the kit.The expression of α-SMA,fibronectin and TGF-β1 in lung tissues were detected by Western blotting.Results Compared with the control group,the lung injury score and inflammation score of the OVA group were increased,the total number of white blood cells,e-osinophils,lymphocytes and neutrophils were increased,the contents of total IgE and OVA-specific IgE were increased,and the levels of IL-12,IFN-γ,IL-4 and IL-13 were increased.The collagen deposition in bronchial subepithelium was significantly increased,the content of HYP was increased,and the expression levels of α-SMA,fibronectin and TGF-β1 were increased(all P<0.05).Compared with the OVA group,lung tissue injury,lung injury score and inflammation score,total white blood cell count,eosinophils,lymphocytes and neutrophils,total IgE and OVA-specific IgE content,IL-12 and IFN-y levels were reduced in the OVA+miR-126 inhibitor group.The levels of IL-4 and IL-13 increased,the subepithelial collagen deposition decreased,the content of HYP decreased,the expression levels of α-SMA,Fibronectin and TGF-β1 decreased,and the differences were statistically significant(all P<0.05).Conclusion miR-126 inhibitor can improve the immune balance,pulmonary fibrosis and collagen deposition induced by ovoalbumin in asthmatic mice.
