摘 要:目的探讨羊耳菊水提物(water extract of Inula cappa)保护前交叉韧带切断术诱导的骨关节炎大鼠的可能机制.方法构建骨关节炎大鼠模型,术后给予6、12、24 g/kg羊耳菊水提物灌胃.28 d后,检测骨体积分数(BV/TV)、特定骨表面积(BSA/BV)、骨小梁厚度(Tb.Th)、骨小梁分离度(Tb.Sp)、肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、IL-10水平;统计骨小梁及软骨含量,检测软骨组织中Osx、Col1a1、Dlx2基因表达以及WOSCAR、TRAP蛋白表达.结果中、高剂量羊耳菊水提物干预后,能够提高大鼠的BV/TV、Tb.Th水平、骨小梁、软骨含量、Osx、Dlx2、Col1α1相对表达水平(P<0.05),降低BSA/BV、Tb.Sp、TNF-α、IL-1β、IL-10、TAPR染色阳性率、OSCAR、TRAP蛋白表达水平(P<0.05).结论羊耳菊水提物可能通过抑制炎性应激反应降低软骨组织处的免疫反应,恢复成/破骨细胞的功能平衡,改善软骨退变程度,从而保护骨关节炎大鼠.Objective To explore the possible mechanism of water extract of Inula cappa(CWE)on protecting rats with osteoarthritis(OA)induced by anterior cruciate ligament transaction(ACLT).Methods OA rat models were constructed.They were intragastrically administrated with CWE(6,12,24 g/kg)after ACLT.After 28 days,bone volume fraction(BV/TV),surface area of specific bone(BSA/BV),thickness of bone trabecula(Tb.Th),bone trabecular separation(Tb.Sp),and levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and IL-10 were detected.The contents of bone trabecula and cartilages were statistically analyzed.The expression levels of Osx,Collal and Dlx2 genes in cartilage tissues,OSCAR and TRAP proteins were detected.Results After intervention with medium-dose and high-dose CWE,BV/TV and Tb.Th levels,contents of bone trabecula and cartilages,and relative expression levels of Osx,Dlx2 and Collal were significantly increased(all P<0.05),and BSA/BV,Tb.Sp,TNF-α、IL-1β,IL-10,positive rate of TAPR staining,and expression levels of OSCAR and TRAP proteins were significantly decreased(all P<0.05).Conclusion CWE may reduce immune response in cartilage tissues by inhibiting inflammatory stress response,recover the functional balance of osteoblasts and osteoclasts,and improve cartilages degeneration,thus protecting OA rats.
