摘 要:目的探索白毛藤多糖对人甲状腺乳头状癌TPC-1细胞的影响及其调控机制.方法在体外用不同浓度(0、0.4、0.8、1.6 mg/ml)的白毛藤多糖处理人甲状腺乳头状癌TPC-1细胞,白毛藤多糖对TPC-1细胞增殖活性的影响采用MTT法检测,对TPC-1细胞凋亡的影响采用Hoechest 33258染色法检测,同时通过Western印迹检测白毛藤多糖对TPC-1细胞JNK蛋白表达及JNK磷酸化的影响.结果MTT结果显示,与空白组相比,不同浓度白毛藤多糖处理后TPC-1细胞增殖抑制率显著升高,差异有统计学意义(P<0.05);且白毛藤多糖浓度越高、处理时间越长,对TPC-1细胞增殖活性的抑制作用越明显(P<0.05).Hoechest 33258染色结果显示,与空白组相比,不同浓度白毛藤多糖处理后TPC-1细胞凋亡率显著升高,且呈浓度依赖(P<0.05);同时,Western印迹结果表明,与空白组相比,不同浓度白毛藤多糖处理后TPC-1细胞中Bcl-2/Bax比率凋亡率显著降低,且呈浓度依赖(P<0.05).经不同浓度白毛藤多糖处理后TPC-1细胞中,JNK及p-JNK蛋白表达均显著降低,且随着白毛藤多糖浓度的增加,其对JNK及p-JNK蛋白表达的抑制逐渐增强(P<0.05);此外,白毛藤多糖对p-JNK的抑制更为显著,随着白毛藤多糖浓度的增加,p-JNK/JNK比值逐渐下降(P<0.05).结论白毛藤多糖对人甲状腺乳头状癌TPC-1细胞的增殖具有抑制作用,其机制可能与凋亡通路中活化Bax蛋白、抑制Bcl-2蛋白及抑制JNK磷酸化有关.Objective To explore the effects of solanum lyratum thunb(SLT)polysaccharide on the proliferation of human thyroid papillary carcinoma(TPC-1)cells and the regulatory mechanism.Methods The human TPC-1 cells were treated with different concentrations of SLT polysaccharide(0,0.4,0.8,1.6 mg/ml)in vitro.The proliferation of TPC-1 cells was detected by MTT,the apoptosis of TPC-1 cells by Hoechest 33258 staining and the expression and phosphorylation of Jun N-terminal kinase(JNK)by Western blotting.Results MTT results showed that the growth of TPC-1 cells was significantly inhibited after treatment with different concentrations of SLT polysaccharide when compared with that in blank group(P<0.05).This inhibitory effect was time-and concentration-dependent(P<0.05).Hoechest 33258 staining showed that the apoptosis rate of TPC-1 cells was significantly increased in SLT polysaccharide groups relative to blank group,which was also in a concentration-dependent manner(P<0.05).Western blot results showed the Bcl-2/Bax in TPC-1 cells was concentration-dependently decreased in SLT polysaccharide groups as compared with the blank group(P<0.05).After treatment with different concentrations of SLT polysaccharide,the expression levels of JNK and p-JNK proteins in TPC-1 cells were significantly decreased(P<0.05).The inhibitory effects of SLT polysaccharide on p-JNK were more significant.With the increase of the concentration of SLT polysaccharide,p-JNK/JNK was gradually decreased(P<0.05).Conclusion SLT polysaccharide could significantly suppress the proliferation of human TPC-1 cells.The mechanism may be related to the activation of Bax protein,and the inhibition of Bcl-2 protein and JNK phosphorylation in apoptosis pathway.
