摘 要:目的:应用网络药理学方法,探讨平消片治疗胃癌的作用机制。方法:使用ETCM数据库收集平消片的作用靶点,使用DisGeNET数据库收集胃癌疾病的靶点,取交集获得平消片治疗胃癌的潜在靶点,对潜在靶点进行GO、KEGG富集分析,构建蛋白互作网络,获得核心靶点及转录因子;分析肿瘤相关免疫细胞浸润情况;评估作用靶点的临床意义及预后价值。结果:获得平消片治疗胃癌的潜在靶点38个,富集分析结果表明平消片可以通过调节脂质代谢过程、炎性反应、免疫、花生四烯酸代谢、SUMO化修饰等多种生物途径发挥抗胃癌功能,也可作用于PTGS2、AKT1、ESR1、NR3C1、AHR等核心靶点并影响胃癌患者预后。结论:平消片可以通过多途径、多靶点发挥抗胃癌作用,为进一步研究提供了理论依据和线索。Objective:To explore the mechanism of Pingxiao tablets in the treatment of gastric cancer by network pharmacology.Methods:The ETCM database was used to collect the targets of Pingxiao tablet,and the DisGeNET database was used to collect the targets of gastric cancer.GO and KEGG enrichment analyses were performed based on the potential targets.The protein interaction network was constructed,and hub targets and transcription factors were obtained.The infiltration of tumor related immune cells was analyzed.The clinical significance and prognostic value of the targets were evaluated.Results:38 potential targets of Pingxiao tablet in the treatment of gastric cancer were obtained.The results of enrichment analysis showed that Pingxiao tablet can exert anti-gastric cancer function by influencing lipid metabolic process,inflammatory response,immunity,arachidonic acid metabolism,SUMOylation and other biological pathways.It can also act on hub targets such as PTGS2,AKT1,ESR1,NR3 C1 and AHR,and affect the prognosis of patients with gastric cancer.Conclusion:Based on the method of network pharmacology,the study found that Pingxiao tablet can play an anti-gastric cancer role through multiple pathways and targets,which provides theoretical basis and clues for further research.
