摘 要:目的:探讨促红细胞生成素(EPO)对急性肾损伤大鼠肾小管细胞凋亡和MAPKAPK-2水平的影响。方法:30只SD雄性大鼠分为假手术组、模型组、EPO治疗组,假手术组仅分离肾被膜后逐层关闭腹腔;模型组采用缺血再灌注法建立急性肾损伤模型;EPO治疗组为急性肾损伤大鼠腹腔注射EPO 50 U/kg,每周3次,共6周,其余大鼠注射等体积的生理盐水。ELISA法检测各组大鼠肾功能指标改变情况,H-E染色观察各组大鼠肾组织结构改变情况,RT-PCR和免疫印迹检测肾组织p-MAPKAPK-2 mRNA和蛋白表达水平,流式细胞术检测肾小管细胞凋亡率。结果:与假手术组相比,模型组大鼠肾功能降低;与模型组相比,EPO治疗组肾功能好转,差异均有统计学意义;假手术组大鼠肾组织未见水肿及炎症细胞浸润,结构完整。模型组大鼠肾组织大量水肿,有炎症细胞浸润且肾间质见散在出血点,EPO治疗组肾组织明显改善,水肿缓解;与假手术相比,模型组MAPKAPK-2 mRNA水平、蛋白水平均升高,肾小球细胞凋亡率升高;与模型组相比,EPO治疗组上述指标均降低,差异均有统计学意义。结论:EPO能抑制急性肾损伤大鼠肾小管细胞凋亡,降低p-MAPKAPK-2的水平,减轻急性肾损伤程度。Objective:To investigate the effect of erythropoietin(EPO)on renal tubular cell apoptosis and mapkapk-2 level in rats with acute kidney injury(AKI).Methods:Thirty male Sprague Dawley rats were divided into sham operation group,model group and EPO treatment group.The AKI model was established by ischemia/perfusion.Animals in EPO treatment group were treated with abdominal injection of 50 U/kg EPO three time a week for six weeks,while rats in sham and model groups received abdominal adminstration of normal saline.The changes of renal function were detected by ELISA.The changes of renal tissue structure were observed by H-E staining.The expression of p-MAPKAPK-2 mRNA was detected by RT-PCR.The protein level of p-MAPKAPK-2 was detected by Western blot.The apoptosis rate of renal tubular cells was detected by flow cytometry.Results:Compared with the sham operation group,the renal function of the model group decreased.Compared with the model group,the EPO treatment group increased,there was a significant difference;The staining results showed that there was no edema and inflammatory infiltration in the renal tissue of the sham operation group,and the structure was complete.The results of Western blotting and PCR showed that compared with sham operation,the protein and mRNA levels of MAPKAPK-2 in the model group were increased,and the above indexes in the EPO treatment group were decreased.The results of flow cytometry showed that compared with the sham operation group,the apoptosis rate of glomerular cells in the model group was increased;with a significant difference compared with the model group,the apoptosis rate in the EPO treatment group was significantly decreased.Conclusion:EPO can inhibit the apoptosis of renal tubular cells in rats with AKI and reduce the level of p-MAPKAPK-2 and acute kidney injury.
