摘  要:目的探讨跨膜水通道蛋白(Aquaporin,AQP)1、7与小鼠肾小管发生发育的关系,为研究和治疗相关性肾脏疾病提供实验依据。方法动物选用胚龄(E)12、14、17、18 d及生后日龄(P)1、3、7、14、24、40、70 d的小鼠;免疫组织化学术观察AQP1、AQP7在小鼠肾小管中的表达,体视学方法测量二者阳性表达的面密度值;蛋白印迹检测AQP1、AQP7在发育各阶段肾组织中的含量变化。结果免疫组织化学显示在E14 d的肾小管中AQP1、AQP7均有阳性表达,之后AQP1主要定位在近端小管、髓袢降支细段的管腔膜和侧基底膜,AQP7则主要定位在皮质和外髓部近直小管的刷状缘;体视学结果显示,随着肾脏发育,肾小管中表达的AQP1、AQP7面密度值均逐渐升高,然后趋于平稳;蛋白印迹显示AQP1在肾脏的表达量在P7 d达到高峰后趋于稳定;AQP7则是P24 d达到高峰后趋于稳定。结论AQP1、AQP7在肾小管的发育过程中存在时空性表达,对肾小管重吸收水、甘油功能的形成起重要作用。Objective To investigate the relationship between AQP1,AOP7 and renal tubules development,and provide theoretical basis for research and treatment of kidney diseases.Methods Animals were selected from embryonic days(E)12,14,17,18 and postnatal days(P)1,3,7,14,24,40,70.The expression of AQP1 and AQP7 were examined by immunohistochemistry method in the renal tubules.The surface area density values of AQP1 and AOP7 positive expressions were measured by stereological method,while the contents of AQP1 and AOP7 in the kidney of mice were examined by Western blot.Results There were AQP1 and AQP7 positive staining in developing renal tubules at E14 day by immunohistochemical analysis.AQP1 localized at the apical and basolateral surfaces of the proximal convoluted tubules,the descending thin limbs of the loop of Henle;AQP7 localized along the brush border of the proximal straight tubules(S3 segment)where the cortex and outer stripe of the outer medulla developed after P14 d.The results of stereology showed that the surface area density values of AQP1and AQP7 were increased gradually and then remained stable during tubule development.Western blot indicated that AQP1 expression reached the peak at P7 d and then remained stable.AQP7 expression reached the peak at P24 d and then remained stable.Conclusion The expressions of AQP1 and AQP7 in the developing renal tubules of mice show a chronological and spatial sequence,which plays an important role in the development and maturation of renal tubule for reabsorption of water and glycerol.
