摘 要:目的:研究RBM10,caspase-9和eNOS在早中期胚胎蜕膜组织中的表达及意义。方法:通过培育孕鼠获得6.5~9.5 d胚胎蜕膜组织,制作组织蜡块切片。采用免疫组化技术检测RBM10、caspase-9和eNOS在早中期蜕膜组织中的表达,应用ImageProPlus图像处理软件分析图像。结果:(1)RBM10表达于蜕膜细胞核,合体滋养层细胞核及子宫腺上皮细胞核,caspase-9表达于蜕膜腺上皮细胞质及部分基质细胞胞质。(2)Caspase-9在孕鼠8.5 d的蜕膜组织中表达高于6.5 d、9.5 d(P<0.05);RBM10在孕鼠8.5 d的子宫蜕膜中表达高于6.5 d,且在8.5 d时达高峰后下降,9.5 d时的表达低于8.5 d(P<0.05);eNOS在孕鼠蜕膜化的6.5 d、8.5 d和9.5 d呈弱阳性表达,且8.5 d时的表达水平高于9.5 d(P<0.05)。结论:caspase-9、RBM10和eNOS可能参与了子宫早中期蜕膜化过程及胚胎组织的生长与分化调控。Objective:To investigate the expressions and significance of RBM10,caspase-9 and eNOS in the decidual tissue of early and mid-term embryos.Methods:The decidual tissue of 6.5~9.5 d embryo was obtained by cultivating pregnant mice,and tissue paraffin section was made.The expression level of RBM10,caspase-9 and eNOS in early and mid-term decidual tissues was detected by immunohistochemical technique and the images were analyzed by the processing software ImageProPlus.Results:(1)The expression of RBM10 was concentrated in the decidual nucleus,syncytiotrophoblast nucleus and uterine glandular cell nucleus,and caspase-9 was mainly expressed in the epithelial cytoplasm of decidual glandand part of stromal cytoplasm.(2)the expression level of Caspase-9 in decidual tissue of 8.5 d pregnant rats was higher than that of 6.5 d and 9.5 d pregnant rats(P<0.05);the expression of RBM10 in decidua tissue of 8.5 d pregnant was higher than that of 6.5 d pregnant rats,and the expression peaked at 8.5 d and then decreased,and it was lower at 9.5 d than 8.5 d(P<0.01);and the expression of eNOS was weakly positive at 6.5 d,8.5 d and 9.5 d,and its expression level at 8.5 d was higher than at 9.5 d.Conclusion:Caspase-9,RBM10 and eNOS may be involved in the early and middle-term uterine decidualization and the regulation of growth and differentiation of embryonic tissue.
