摘 要:目的探讨瑞芬太尼对缺氧/复氧诱导的心肌细胞损伤影响及其对miR-574-3p的调控作用。方法体外培养大鼠H9C2心肌细胞为对照组,缺氧/复氧诱导H9C2细胞建立心肌缺血再灌注损伤模型为模型组,用不同剂量瑞芬太尼(10、30、60 ng/ml)处理的细胞为低、中、高剂量组。将anti-miR-con、anti-miR-574-3p转染至H9C2细胞24 h后再进行缺氧/复氧处理并分组,miR-con、miR-574-3p mimics转染至H9C2细胞加入瑞芬太尼60 ng/ml处理后再进行缺氧/复氧处理并分组。检测各组细胞活性、细胞凋亡、丙二醛(MDA)、超氧化物歧化酶(SOD)含量及miR-574-3p、CyclinD1、Cleaved-caspase-3蛋白表达水平。结果与对照组比较,模型组细胞活性、SOD含量、CyclinD1蛋白表达水平降低,细胞凋亡率、MDA含量、Cleaved-caspase-3、miR-574-3p蛋白表达水平增高,差异有统计学意义(P<0.05)。与模型组比较,低、中、高剂量组细胞活性、SOD含量、CyclinD1蛋白表达水平升高,细胞凋亡率、MDA含量、Cleaved-caspase-3、miR-574-3p蛋白表达水平降低,且随着瑞芬太尼剂量的增加变化更显著,差异有统计学意义(P<0.05)。与anti-miR-con组比较,anti-miR-574-3p组细胞活性、CyclinD1蛋白表达水平、SOD含量升高,细胞凋亡率、Cleaved-caspase-3蛋白水平、MDA含量降低,差异有统计学意义(P<0.05)。与高剂量+miR-con组比较,高剂量+miR-574-3p组细胞活性、CyclinD1蛋白表达水平、SOD含量降低,细胞凋亡率、Cleaved-caspase-3蛋白表达水平、MDA含量升高,差异有统计学意义(P<0.05)。结论瑞芬太尼可通过下调miR-574-3p的表达而增强细胞活性、抗氧化能力及抑制细胞凋亡,进而减轻缺氧/复氧诱导的心肌细胞损伤。Objective To explore effect of Remifentanil on cardiomyocyte damage induced by hypoxia/reoxygenation(H/R)and its regulatory effect on miR-574-3p.Methods Rats'cardiomyocytes H9C2 by culture in vitro were used as control group,while myocardial ischemia-reperfusion injury models which were established by H/R-induced H9C2 cells were used as model group.Cells were treated with different doses of Remifentanil(10,30 and 60 ng/ml)and divided into low-,moderate-and high-dose groups.Anti-miR-con and anti-miR-574-3p were transfected into H9C2 cells for 24 h before H/R treatment and grouped.The miR-con and miR-574-3p mimics were transfected into H9C2 cells,and then Remifentanil was added to the cells before H/R treatment and grouped.Cell viability and apoptosis,contents of malondialdehyde(MDA)and superoxide dismutase(SOD),and expressions of miR-574-3p,CyclinD1 and Cleaved-caspase-3 proteins were detected in all groups.Results Compared with those in control group,values of cell activity,SOD content and CyclinD1 protein expression were significantly decreased,while values of cell apoptosis rate,MDA content,and protein expressions of Cleaved caspase 3 and anti miR 5743p were significantly increased in model group(P<0.05).Compared with those in model group,values of cell activity,SOD content and CyclinD1 protein expression were significantly increased,whereas values of apoptosis rate,MDA content,and protein expressions of Cleaved caspase 3 and anti miR 5743p were significantly decreased in low-,moderate-,and high-dose groups,which were more significant with increasing dose of Remifentanil(P<0.05).Compared with those in anti-miR-con group,values of cell activity,CyclinD1 protein expression and SOD content were significantly decreased,while values of cell apoptosis rate,Cleaved-caspase-3 protein expression and MDA content were significantly increased in anti-miR-574-3p group(P<0.05).Compared with those in high-dose+miR-con group,values of cell viability,CyclinD1 protein expression and SOD content were significantly increased,while
