摘  要:目的:基于网络药理学探讨康莱特注射液治疗肝癌的作用机制。方法:通过TCMSP数据库筛选康莱特注射液中的有效成分和其作用靶点,通过GeneCards数据库获取筛选肝癌相关的疾病靶点,整合筛选药物靶点和疾病靶点交集的核心靶点,利用Cytoscape 3.6.1软件构建“药物-成分-靶点-疾病”网络。结合String数据库构建成分靶点-疾病靶点蛋白相互作用全局网络,并通过生物学手段对核心靶点进行生物学功能分析和通路富集分析。结果:经筛选获得9个活性成分,对应41个靶点,其中与肝癌相关有25个重叠靶点,对该25个靶点进行了GO富集分析,KEGG富集分析得到22条信号通路。结论:初步验证了康莱特注射液多成分和多靶点的特点,预测了康莱特注射液治疗肝癌可能的作用机制,为其实验研究提供理论依据。Objective To explore the mechanism of Kanglaite injection in the treatment of liver cancer based on network pharmacology.Methods TCMSP database was used to screen the effective components and their targets in Kanglaite injec⁃tion,and GeneCards database was used to obtain the disease targets related to liver cancer.The key targets of drug targets and disease targets were integrated and screened.Cytoscape 3.6.1 software was used to construct the"drug-ingredient-tar⁃get-disease"network.The global network of interaction between component targets and disease target proteins was construc⁃ted with string database,and the biological function and pathway enrichment of core targets were analyzed by bioinformatics.Results The global network of interaction between component targets and disease target proteins was constructed with string database,and the biological function and pathway enrichment of core targets were analyzed by bioinformatics.Conclusion The multi-component and multi-target characteristics of Kanglaite injection were preliminarily verified,and the possible mechanism of Kanglaite injection in treating liver cancer was predicted,which provided a theoretical basis for its experi⁃mental research.
