摘 要:以根结线虫病样、番茄品种及组合为试材,采集包头市根结线虫暴发地一个根结线虫病样,进行形态学和分子生物学鉴定;利用PCR技术对24个番茄品种及组合进行Mi-1基因扩增筛选。结果表明,感染内蒙古包头市番茄的根结线虫为南方根结线虫;筛选出3个纯合抗根结线虫番茄组合。抗感品种及组合均产生750 bp的特异片段,纯合抗病基因的番茄组合存在TaqⅠ酶切位点,酶切后产生了570和160 bp特异性片段,为防治当地番茄根结线虫及培育抗病品种积累材料。Taking the root knot nematode samples and tomato varieties and combinations as test materials,a root knot nematode disease sample from the root knot nematode outbreak area in Baotou City was collected for morphological and molecular biological identification.The Mi-1 gene amplification and screening of 24 tomato varieties and combinations were carried out by PCR technology.The results showed that the root knot nematode infected tomato in Baotou City of Inner Mongolia was the southern root knot line,three homozygous tomato combinations with resistance to root knot nematode were screened out.The resistant varieties and combinations all produced 750 bp specific fragments.The Taq I restriction site existed in the homozygous resistant tomato combinations,and 570 and 160 bp specific fragments were produced after enzyme digestion,which were used to accumulate materials for the control of local tomato root knot nematode and the cultivation of disease resistant varieties.
