摘  要:目的研究鸡血藤对骨质疏松性骨折大鼠内固定术后恢复的作用及机制。方法60只大鼠随机分为对照组、模型组、鸡血藤低、中、高剂量组5组,每组12只。除对照组外,其余大鼠采用卵巢摘除法建立骨质疏松大鼠模型,然后进行开放性右侧股骨中段骨折,并用克氏针行髓内固定术治疗,术后第二天开始,鸡血藤低、中、高剂量组根据大鼠体重分别按照50 mg/kg、100 mg/kg、200 mg/kg灌胃给予大鼠鸡血藤提取物,对照组及模型组灌胃给予等量生理盐水,所有组别每天给药1次,连续给药8周后,测定大鼠右侧股骨骨折端骨密度、骨生物力学,苏木精-伊红染色进行组织病理学观察,Western blot检测大鼠股骨骨折组织中LRP5、β-catenin、Runx2蛋白水平。结果与对照组相比,模型组及鸡血藤低、中、高剂量组大鼠全身骨密度均显著降低(P<0.05),连续给药8周后,与模型组相比,鸡血藤各剂量组大鼠股骨骨折端骨密度、最大载荷和抗弯强度均显著升高(P<0.05),组织病理学变化明显改善,鸡血藤各剂量组大鼠股骨骨折组织中LRP5、β-catenin、Runx2蛋白水平均显著升高(P<0.05),且各项指标的变化与鸡血藤的剂量表现出依赖性。结论鸡血藤能够促进骨质疏松性骨折大鼠内固定术治疗后骨密度、骨生物力学等的恢复,其作用机制可能与调节Wnt/β-catenin信号通路有关。Objective To study the effect and mechanism of Caulis spatholobi on the recovery of osteoporotic fracture in rats after internal fixation.Methods Sixty rats were randomly divided into control group,model group,low,middle and high dose groups.Except for the control group,the osteoporotic rat model was established by ovariectomy.The whole body bone mineral density was measured by dual energy X-ray absorptiometry to evaluate whether the rat model was established,and then the open fracture of the middle part of the right femur was used.Intramedullary fixation with Kirschner needle was performed from the second day after operation.According to the body weight of rats,the rats in the low,middle and high dose groups were given intragastric administration of the extract according to 50mg/kg,100mg/kg and 200mg/kg respectively,while the control group and model group were given the same amount of normal saline.All groups were given once a day.After continuous administration for 8 weeks,the bone mineral density and bone biomechanics of the fracture end of the right femur were measured,and the histopathology was observed by hematoxylin-eosin staining.The protein levels of LRP5,β-catenin and Runx2 in rat femoral fracture tissue were detected by Western blot.Results Compared with the control group,the whole body bone mineral density of rats in the model group and the low,middle and high dose groups decreased significantly(P<0.05).After continuous administration for 8 weeks,compared with the model group,the bone mineral density,maximum load and bending strength of the femur fracture end of rats in each dose group were significantly increased(P<0.05),and the histopathological changes were significantly improved.The protein levels of LRP5,β-catenin and Runx2 in the femur fracture tissue of rats in each dose group were significantly increased(P<0.05),and the changes of each index were dependent on the dose of Caulis spatholobi.Conclusion Caulis spatholobi can promote the recovery of bone mineral density and bone biomechanics afte
