Objective To investigate the key protein and m RNA levels of Wnt/β-Catenin signal transduction pathway in the primary keloid fibroblasts, and their roles in formation mechanism of keloid. Methods Primary fibroblasts were cultured by tissue culture. The protein and m RNA levels of Wnt5 a,β-Catenin,GSK-3β,p-GSK-3β of Wnt/β-Catenin signal transduction pathway in normal skin and the keloid fibroblasts were respectively detected by Western blot and RT-PCR.Data were analyzed by SPSS18.0 software. Results Compared with normal skin, the protein levels of Wnt5 a,β-Catenin,p-GSK-3β and m RNA of Wnt5 a,β-Catenin in keloid fibroblasts were increased(P<0.05),but the expression levels of GSK-3βprotein and m RNA did not significantly change. Conclusions In the process of keloid formation, both m RNA and protein levels of Wnt5 a and β-Catenin up-regulated.Phosphorylation of GSK-3β protein led to the upregulation of p-GSK-3β,which may play an important role in the formation of keloid.
