摘  要:质子泵抑制剂是一种酸抑制药,广泛应用于胃食管反流病、胃十二指肠溃疡等消化性疾病的治疗,与非甾体抗炎药合并使用可显著减少非甾体抗炎药的消化道不良反应。近期研究表明,长期使用质子泵抑制剂有导致骨折的风险,且骨折的风险与质子泵抑制剂使用的剂量及用药时间有关。目前研究显示,质子泵抑制剂增加骨折风险相关性时出现2种研究结果,一种是质子泵抑制剂会导致骨质疏松,另一种是导致类石骨症而增加骨脆性。本文阐述质子泵抑制剂与骨折风险的相关性及其机制,为其临床应用提供依据。Proton pump inhibitor is a kind of acid suppressive drug widely used in gastro-esophageal reflux disease,gastroduodenal ulcer and other digestive diseases.The combination of PPIs with NSAIDs can significantly reduce the adverse reactions and side effects of NSAIDs.Recent studies have shown that the long-term use of PPI has the risk of fracture,and the risk of fracture is related to the dosage and time of using PPIs.At present,two similar results have been found in the study of the correlation between proton pump inhibitors and fracture risk.One is that proton pump inhibitors can lead to osteoporosis,and the other is that they can lead to osteoid and increase bone fragility.In this paper,the relationship of proton pump inhibitors with fracture risk and its mechanism are discussed,which provides basis for its future clinical application.
