摘 要:杜鹃花(Rhododendron spp.)是北温带植物区系的重要类群和世界著名的观赏花卉。本研究利用11个ISSR标记在32个杜鹃花品种内检测到44个多态性位点,品种内多样性丰富,平均Na为1.978,Ne为1.407,多样性指数h和I分别为0.263和0.414。聚类分析将32个品种分为2簇,第一簇含有18个红色、粉红色、紫色花系的品种,第二簇为14个白色和有条纹镶嵌的品种。筛选出10个品种特异性条带,引物844对‘吉见戈玉’和‘梅花茸’在350 bp处分别有一条明显的特异性带;引物846对‘外国红’在175 bp处有一条明显的特异性带;引物847对‘紫鹃’在750 bp处有一条明显的特异性带。本研究构建的DNA指纹图谱为杜鹃花品种鉴定提供了参考。Rhododendron,an important species of north temperate zone,is famous ornamental flowers all over the world.In this research,44 polymorphic loci were detected in 32 Rhododendron cultivars using 11 ISSR markers.High genetic diversity existed among these cultivars,as average Na and Ne was 1.978 and 1.407,respectively.In addition,the diversity index h and I were 0.263 and 0.414,respectively.Cluster analysis divided these 32 varieties into two clusters(clade I and clade II).Clade I contained 18 varieties with red,pink and purple flowers,while clade II was formed by 14 varieties with white and stripe inlay.In total,ten variety-specific bands were screened out.In particular,primer’844’gave out an obvious specific band at 350 bp in cultivars’Jijiangeyu’and’Meihuarong’.Moreover,a 175 bp specific band was amplified by primer’846’in cultivar’foreign red’.A typical band of 750 bp was amplified by primer’847’in cultivar’Zijuan’.The DNA fingerprint constructed in this study will provide critical reference for the identification of Rhododendron varieties.
