摘 要:以臭参为试验材料,经过试样制备、提取纯化、测定等步骤,分析臭参皂苷含量,重点研究了试样制备方式、皂苷元溶解度、洗脱乙醇浓度、回收率及精密度等。结果表明:采用水浴加热方式提取臭参皂苷的效果优于超声方式;薯蓣皂苷元、臭参总皂苷都可溶于70%乙醇;选择无水乙醇作为洗脱剂;臭参地下、地上部分的总皂苷含量分别为1.08%、1.18%;回收率和精密度都较理想。检测方法适用于臭参总皂苷及其他皂苷元含量较多材料的检测;臭参地上部分有较高的开发利用价值。Using Codonopsis bulleyana as the test material,the content of saponin was analyzed after sample preparation,extraction and purification,and determination.It focused on the sample preparation method,the solubility of sapogenin,the concentration of elution ethanol,the recovery rate and the precision.The results showed that using water bath heating method to extract saponins of Codonopsis bulleyana was better than ultrasonic method;diosgenin and total saponins of Codonopsis bulleyana were soluble in 70%ethanol;selecting absolute ethanol as the eluent;total saponins of aboveground and underground parts of Codonopsis bulleyana were 1.08%,1.18%,respectively;the recovery rate and the precision were both ideal.The method is suitable for the determination of total saponins of Codonopsis bulleyana and other materials with more saponins.The aboveground part of Codonopsis bulleyana has high development and utilization value.
