不同种群牛膝菊细胞POD酶的低温响应

摘  要:采用酶定位技术,在超微结构水平上,分析牛膝菊不同种群的POD酶细胞化学定位和低温响应。结果表明:自然条件下,敏感、抗性种群POD的酶活一致,定位位点相同,均在细胞壁。2℃低温处理后,2个种群间POD酶的定位量和定位位点均发生明显变化。其中抗性种群在低温处理8h时酶活定位量最多,且在细胞膜、细胞间隙也出现明显定位点;处理12h后细胞结构完好,细胞壁上仍显示酶活性。敏感种群的POD酶活定位量逐渐减少,且只出现在细胞壁上;当低温处理8h时已没有酶活性,细胞膜受害,发生质壁分离;低温处理12h时整个叶片软化。POD酶活性与牛膝菊不同抗性种群在低温下的适应能力相关。By using the enzyme localization technique,we analyzed POD cytochemical localization and low temperature response of two populations of Galinsoga parviflora.The results showed that POD activity and the localization of two populations were the same without temperature treatment.POD activity and the localization of them were different at 2℃.POD activity of resistant population was the strongest after 8 h with POD localized on the intercellular space and plasma membranes.The cell structure was intact after 12 h.However,POD activity of sensitive populations gradually decreased and POD was localized only on the cell wall.POD activity was inactivated and the cell membrane was destroyed after 8 h.POD activity of G.parviflora was associated with low temperature adaptability.
