摘  要:目的:探索马甲子提取物对宫颈癌细胞增殖、凋亡的影响及其分子机制。方法:分别使用0,25,50,75μg·ml^(-1)马甲子提取物处理宫颈癌细胞SiHa,噻唑蓝(MTT)检测细胞增殖,流式细胞术检测细胞凋亡,蛋白质印迹法(Western blot)检测细胞周期蛋白D1(Cyclin D1)、P21、B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax)表达,实时荧光定量PCR(qPCR)检测长链非编码RNA(lncRNA)人类白细胞抗原复合体18(HCG18)表达。在细胞中转染si-HCG18,观察抑制HCG18对细胞SiHa增殖、凋亡的影响。在细胞中转染pc DNA 3.1-HCG18,并使用75μg·ml^(-1)马甲子提取物进行处理,探讨过表达HCG18对马甲子提取物诱导的SiHa增殖、凋亡的影响。结果:与0μg·ml^(-1)马甲子提取物组比较,25,50,75μg·ml^(-1)马甲子提取物显著提高SiHa细胞的增殖抑制率、凋亡率、P21、Bax蛋白水平,显著减少Cyclin D1、Bcl-2蛋白表达量及HCG18表达量,且呈浓度依赖性(P<0.05)。抑制HCG18显著减少SiHa细胞HCG18表达量及Cyclin D1、Bcl-2蛋白表达量,显著提高P21、Bax蛋白水平、细胞增殖抑制率、凋亡率(P<0.05)。过表达HCG18能逆转马甲子提取物抑制细胞SiHa增殖、Cyclin D1、Bcl-2蛋白表达,以及促进细胞SiHa凋亡、P21、Bax蛋白表达的作用。结论:马甲子提取物通过调控lnc RNA HCG18表达抑制宫颈癌细胞增殖和诱导细胞凋亡。Objective:To explore the effects and molecular mechanism of the extracts from Paliurus ramosissimuson in the proliferation and apoptosis of cervical cancer cells.Methods:Cervical cancer cells SiHa were respectively treated with 0,25,50 and 75μg·ml^(-1) extracts from P.ramosissimuson,and the cell proliferation was detected by MTT assay,and the apoptosis was determined by flow cytometry.The expressions of Cyclin D1,P21,Bax and Bcl-2 were detected by Western blot.q PCR was applied to detect the expression of lnc RNA HCG18.Si-HCG18 was transfected into cells,and the effects of inhibiting HCG18 on the proliferation and apoptosis of SiHa cells were observed.pc DNA3.1-HCG18 was transfected into cells and treated with 75μg·ml^(-1) extracts from P.ramosissimuson to investigate the effects of overexpression of HCG18 on the proliferation and apoptosis of SiHa induced by Jatropha extracts.Results:Compared with the 0μg·ml^(-1) extracts from P.ramosissimuson,25,50 and 75μg·ml^(-1) extracts from P.ramosissimuson significantly increased the proliferation inhibition rate,apoptosis rate,P21 and Bax protein levels of SiHa cells,and significantly reduced Cyclin D1 and Bcl-2 protein expression levels and HCG18 expression level in a concentration-dependent manner(P<0.05).Inhibition of HCG18 significantly reduced the expressions of Cyclin D1 and Bcl-2 protein in SiHa cells,while significantly increased P21,Bax protein,cell proliferation inhibition rate and apoptosis rate(P<0.05).Overexpression of HCG18 reversed the inhibitory effects of the extracts from P.ramosissimuson on the cell SiHa proliferation and Cyclin D1,Bcl-2 protein expressions,and also reversed the promotion effects of apoptosis and P21,Bax protein expressions.Conclusion:The extracts from P.ramosissimuson inhibit the proliferation and induce the apoptosis of cervical cancer cell by regulating lnc RNA HCG18 expression.
