麻黄细辛附子汤拆方干预CD4^(+)T细胞调控相关细胞因子分泌及STAT6 mRNA表达的研究  被引量:5

摘  要:目的从拆方角度观察麻黄细辛附子汤对CD4^(+)T细胞及其分泌的白细胞介素-4(IL-4)、白细胞介素5(IL-5)、白细胞介素-10(IL-10)、白细胞介素-13(IL-13)、白细胞介素-17A(IL-17A)与转录因子STAT6 mRNA的影响,探讨麻黄细辛附子汤组方配伍机制。方法体外培养C57BL/6小鼠脾脏来源的CD4^(+)T细胞,收集细胞,各组给药干预18 h,收集细胞及上清液,ELISA检测细胞上清液中IL-4的含量,流式细胞术检测细胞上清中IL-5、IL-10、IL-13、IL-17A的含量,实时荧光定量PCR法检测细胞沉淀中STAT6 mRNA的含量。结果1)IL-4:较之对照组,辛附组、细辛组、附子组有降低趋势,但无明显统计学差异(P>0.05),然全方组、麻附组则含量升高(均P<0.05),其中麻黄组显著升高(P<0.001)。2)IL-5:较之对照组,全方组含量有降低趋势,但无明显统计学差异(P>0.05),辛附组含量则升高(P<0.05)。3)IL-17A:与对照组相比,全方组可显著降低其含量(P<0.01),而麻辛组可升高其含量(P<0.05);与全方组相比,麻附组、辛附组、麻黄组、细辛组、附子组含量均显著升高(均P<0.01)。4)各给药组IL-10、IL-13的含量较之对照组无明显统计学差异(P>0.05)。5)STAT6 mRNA:较之对照组,附子组含量降低(P<0.05),尤以辛附组、细辛组明显(P<0.001);全方组、麻附组含量则升高,麻辛组尤为明显(P<0.05或P<0.01)。结论1)麻黄细辛附子汤全方可减少IL-17A,升高IL-4、STAT6 mRNA的表达发挥其免疫调控作用,在IL-5、IL-10、IL-13环节均未能有显著作用。2)麻黄的加入可拮抗细辛、附子的作用,升高IL-4水平,细辛、附子配伍升高IL-5水平,促进T细胞向Th2分化,从而影响对免疫的调节。3)麻黄细辛附子汤及各拆方配伍未能在IL-10与IL-13环节发挥显著免疫调控作用。4)辛附的配伍或者细辛、附子单用可通过降低STAT6 mRNA水平发挥免疫调节作用,而全方、麻附、麻辛配伍则相反,体现不同配伍对STAT6 mRNA的双向调节作用Objective:To observe the effects of Mahuang Xixin Fuzi Decoction on CD4^(+)T cells and secreted IL-4,IL-5,IL-10,IL-13,IL-17A and transcription factor STAT6 mRNA from the perspective of disassembly,and to explore the compatibility mechanism of Mahuang Xixin Fuzi Decoction.Methods:C57BL/6 mouse spleen-de⁃rived CD4^(+)T cells were cultured in vitro,the cells were collected,and each group was administered intervention for 18 hours.The cells and supernatant were collected.ELISA was used to detect the IL-4 content in the cell su⁃pernatant.The content of IL-5,IL-10,IL-13,and IL-17A in the cell supernatant was detected by flow cytometry,and the content of STAT6 mRNA in the cell pellet was detected by real-time fluorescent quantitative PCR.Re⁃sults:1)IL-4:Compared with the control group,the content in the Xinfu group,the Xixin group,and the Fuzi group had a decreasing trend,but there was no significant statistical difference(P>0.05);however,the whole pre⁃scription group and the Mafu group increased(P<0.05),and the Mahuang group increased significantly(P<0.001).2)IL-5:Compared with the control group,the content of the whole prescription group had a decreasing trend,but there was no significant statistical difference(P>0.05),while the content of the Xinfu group increased(P<0.05).3)IL-17A:Compared with the control group,the whole prescription group could significantly reduce its content(P<0.01),while the Maxin group could increase its content(P<0.05);compared with the whole pre⁃scription group,its content in the Mafu group,Xinfu group,Mahuang group,Xixin group,and Fuzi group all in⁃creased significantly(P<0.01).4)The contents of IL-10 and IL-13 in each administration group were not signifi⁃cantly different from the control group(P>0.05).5)STAT6 mRNA:Compared with the control group,the content of the Fuzi group was lower(P<0.05),especially in the Xinfu group and the Xixin group(P<0.001);the content in the whole prescription group and Mafu group increased(P<0.05),especially in the Maxin group(P<0.05 or P<0.01).Co
