摘  要:目的观察疏肝温胆汤对动脉内皮损伤兔肝X受体(LXR)、肿瘤坏死因子-α(TNF-α)的影响。方法将健康雄性新西兰兔随机分为空白对照组、模型对照组、疏肝温胆汤组,通过高脂饲养及小牛血清白蛋白免疫损伤的方法建立新西兰兔动脉内皮损伤模型。造模4周后,空白对照组及模型对照组予0.9%氯化钠注射液灌胃,疏肝温胆汤组予等量的疏肝温胆汤灌胃,药物干预时间为4周。于实验第8周进行取样送检测,采用苏木素-伊红(HE)染色法检测各组主动脉的病理变化;采用硝酸还原酶法和酶联免疫吸附测定(ELISA)检测各组血清一氧化氮(NO)和内皮素-1(ET-1)的水平;采用实时荧光定量PCR检测主动脉组织中C反应蛋白(CRP)、LXRa、TNF-α的mRNA表达水平。结果动脉内膜病理变化:与空白对照组比较,模型对照组主动脉内膜明显变狭窄,内有大量泡沫细胞生成;与模型对照组比较,疏肝温胆汤组主动脉内膜狭窄程度较小,内有少量泡沫细胞生成。实验室检测指标变化:与空白对照组比较,模型对照组雄兔血浆中NO明显降低(P<0.05),ET-1明显升高(P<0.05),动脉组织中的TNF-α、hs-CRP的mRNA表达水平均明显升高(P<0.05)、LXRαmRNA表达明显降低(P<0.05);与模型对照组比较,疏肝温胆汤组雄兔血浆中NO明显升高(P<0.05),ET-1明显降低(P<0.05),组织的TNF-α、hs-CRP的mRNA表达水平均明显降低(P<0.05),LXRα的mRNA表达明显升高(P<0.05)。结论疏肝温胆汤能改善动脉内皮功能,其作用机制可能与通过调控LXR、TNF-α炎症因子表达有关。Objective:To investigate the effects of Shugan Wendan Decoction on liver X receptor(LXR)and tu⁃mor necrosis factor(TNF-α)in rabbits with arterial endothelial injury.Methods:All healthy male New Zealand rabbits were randomly divided into blank control group,model control group and Shugan Wendan Decoction group.The New Zealand rabbit model of arterial endothelial injury was established by high-fat feeding and immune inju⁃ry of calf serum albumin.After 4 weeks of modeling,rabbits in the blank control group and model control group were given normal saline,and the Shugan Wendan Decoction group were given the same amount of Shugan Wen⁃dan Decoction;the intervention time was 4 weeks.At the 8th week of the experiment,samples were collected and tested.Hematoxylin-Eosin(HE)staining was used to detect the pathological changes of aorta in each group.Se⁃rum nitric oxide(NO)and endothelin-1(ET-1)levels were measured by nitrate reductase method and enzymelinked immunosorbent assay(ELISA).The mRNA expression levels of C-reactive protein(CRP),LXRa and TNFαin aortic tissues were measured by real-time fluorescence quantitative PCR.Results:Pathological changes of ar⁃terial intima:compared with the blank control group,the aortic intima of the model control group was significantly narrowed,and a large number of foam cells were generated.Compared with the model control group,the aortic inti⁃ma of Shugan Wendan Decoction group was relatively wide with a small amount of foam cells.Changes in laborato⁃ry indicators:compared with the blank control group,NO in plasma of male rabbits in the model control group was significantly decreased(P<0.05),ET-1 was significantly increased(P<0.05),mRNA expression levels of TNF-αand Hs-CRP in the arterial tissues were significantly increased(P<0.05),and mRNA expression levels of LXR were significantly decreased(P<0.05).Compared with the model control group,the plasma NO in the Shugan Wendan Decoction group was significantly increased(P<0.05),ET-1 was significantly decreased(P<0.05),the m
