摘 要:目的基于网络药理学探讨参附黄方治疗脓毒症的作用机制。方法通过TCMSP数据库获取红参、附子、大黄的主要化学成分及其靶点;检索GeneCards、OMIM、TTD、Drugbank数据库,得到脓毒症相关靶点;运用R语言得到药物和疾病的交集靶点并绘制韦恩图;应用STRING构建交集靶点相互作用网络图;利用Cyto⁃scape3.7.2软件构建“药物-成分-靶点-疾病”网络图;使用DAVID数据库对靶点进行基因功能(GO)分析和基于京都基因与基因组百科全书(KEGG)通路富集分析。结果参附黄方干预脓毒症的有效活性成分共40个,药物靶点与疾病靶点交集后获得28个共有靶点,核心靶点有TNF、CASP3、TP53、JUN、MYC、PTGS2、IL1B、PPARG等。GO富集分析确定了271个条目;KEGG通路富集分析确定了54条相关信号通路。结论本研究初步揭示了参附黄方调治脓毒症的多靶点、多通路的作用机制,为临床使用及进一步研究提供了一定的理论依据。Objective:To explore the mechanism of ShenFuHuang Formula in the treatment of sepsis based on network pharmacology.Methods:The active components and relevant targets of Red ginseng,Prepared common,Rhubarb root and rhizome were identified from the Traditional Chinese Medicines for Systems Pharmacology Data⁃base and Analysis Platform(TCMSP).Sepsis-related targets were retrieved from GeneCards,OMIM,TTD,and Drugbank databases.R language was used to get drug and disease intersection targets and display the Venn dia⁃gram.The intersection genes were imported into STRING database to construct a protein-protein interaction(PPT)network.The target network diagram“drug-component-target-disease”of ShenFuHuang Formula in treating sepsis was constructed by using Cytoscape 3.7.2.Using DAVID database,the genetic ontology(GO)biological pro⁃cess and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were carried out.Results:There were 40 active ingredients of ShenFuHuang Formula in treating sepsis.The number of targets that intersect with sepsis was 28.TNF,CASP3,TP53,JUN,MYC,PTGS2,IL1B and PPARG may be the key targets of ShenFuHuang Formula in the treatment of sepsis.A total of 271 items were identified by GO enrichment analysis.KEGG pathway enrichment analysis identified 54 related signaling pathways.Conclusions:This network pharma⁃cological study reveals potential mechanisms of multi-target and multi-component ShenFuHuang Formula in the treatment of sepsis,which provides a scientific basis for studying the mechanism.
